In order to demonstrate that high dilutions of histamine are able to inhibit basophil activation in a reproducible fashion, several techniques were used in different research laboratories.
The aim of the study was to investigate the action of histamine dilutions on basophil activation.
Basophil activation was assessed by alcian blue staining, measurement of histamine release and CD63 expression. Study 1 used a blinded multi-centre approach in 4 centres. Study 2, related to the confirmation of the multi-centre study by flow cytometry, was performed independently in 3 laboratories. Study 3 examined the histamine release (one laboratory) and the activity of H(2) receptor antagonists and structural analogues (two laboratories).
High dilutions of histamine (10(-30)-10(-38) M) influence the activation of human basophils measured by alcian blue staining. The degree of inhibition depends on the initial level of anti-IgE induced stimulation, with the greatest inhibitory effects seen at lower levels of stimulation. This multicentre study was confirmed in the three laboratories by using flow cytometry and in one laboratory by histamine release. Inhibition of CD63 expression by histamine high dilutions was reversed by cimetidine (effect observed in two laboratories) and not by ranitidine (one laboratory). Histidine tested in parallel with histamine showed no activity on this model.
In 3 different types of experiment, it has been shown that high dilutions of histamine may indeed exert an effect on basophil activity. This activity observed by staining basophils with alcian blue was confirmed by flow cytometry. Inhibition by histamine was reversed by anti-H2 and was not observed with histidine these results being in favour of the specificity of this effect We are however unable to explain our findings and are reporting them to encourage others to investigate this phenomenon.

    ABSTRACT: Introduction: This study deals with the effects of extremely low doses or high dilutions of pharmacological compounds on in vitro pollen germination of kiwifruit (Actinidia deliciosa). Material and methods: The biological efficacy of As2O3 at the 5th and 45th decimal dilution/succussion level (As 5x and As 45x) was tested on pollen previously stressed with As2O3 150 or 200 μM. The outcome variable was the pollen germination rate, as detected blind after 3 h 30 min by an Axioplan microscope. Results: A directionally consistent recovery of germination percentage was observed in both As 5x and As 45x after stressing with As2O3 150 μM. When pollen was stressed with As2O3 200 μM only as 45x induced a general and significant germination increase. Conclusions: Our results suggest that both treatments might partially remove the inhibitory effect caused by the stressor. Similar effects were observed on the same model using weak extremely low frequency magnetic fields (ELF-MFs) mediated through water. Although preliminary, the findings seem to indicate the in vitro pollen performance as adequate to study the effects of physicochemical subthreshold stimuli (extremely low doses or high dilutions of pharmacological compounds, weak ELF-MFs), mediated through water.

    ABSTRACT: This paper attempts to stimulate the psychological investigation of homeopathy and serially agitated dilutions. The history of homeopathy and serial dilutions is provided in a literature review of selected research areas. Two original illustrative experiments are also presented and discussed. The first examined the effect of serially agitated dilutions of Sevin on the mortality rate of honey bees (Apis mellifera). In a second experiment, the effect of serially agitated dilutions of sucrose on proboscis extension in honey bees was assessed. No differences were found between serially agitated dilutions of pesticides and sucrose compared with dilutions alone. Implications, limitations, and proposed further work are discussed.

    ABSTRACT: Homeopathy is controversial and hotly debated. The conclusions of systematic reviews of randomised controlled trials of homeopathy vary from 'comparable to conventional medicine' to 'no evidence of effects beyond placebo'. It is claimed that homeopathy conflicts with scientific laws and that homoeopaths reject the naturalistic outlook, but no evidence has been cited. We are homeopathic physicians and researchers who do not reject the scientific outlook; we believe that examination of the prior beliefs underlying this enduring stand-off can advance the debate. We show that interpretations of the same set of evidence-for homeopathy and for conventional medicine-can diverge. Prior disbelief in homeopathy is rooted in the perceived implausibility of any conceivable mechanism of action. Using the 'crossword analogy', we demonstrate that plausibility bias impedes assessment of the clinical evidence. Sweeping statements about the scientific impossibility of homeopathy are themselves unscientific: scientific statements must be precise and testable. There is growing evidence that homeopathic preparations can exert biological effects; due consideration of such research would reduce the influence of prior beliefs on the assessment of systematic review evidence.
