Clinical effects of a proprietary combination isoflavone nutritional supplement in menopausal women: a pilot trial.
ABSTRACT As they reach menopause, a majority of women living in Westernized countries experience climacteric symptoms. Hormone replacement therapy (HRT) has been used to remediate these symptoms. Recent studies, however, have suggested that HRT may increase the risk of developing breast cancer and cardiovascular disease (CVD). Therefore, many women are looking for alternative treatment options.
This trial was a pilot study to assess the effect of a nutritional supplement containing isoflavones from kudzu and red clover, along with other targeted nutrients on menopausal symptoms and markers of breast cancer and CVD risk. Twenty-five menopausal women suffering from severe hot flushes and night sweats completed a 12-week intervention using this combination isoflavone nutritional supplement.
We observed a 46% decrease in reported hot flushes, from an average of 9.7 to 5.2 per day. Quality of life, as assessed by the standardized Greene Questionnaire, showed similar improvement. Two markers of CVD risk, the ratio of total cholesterol to high-density lipoprotein (HDL) cholesterol and homocysteine, showed modest improvement. A proposed marker of breast cancer risk, the ratio of 2-hydroxyestrone to 16 alpha-hydroxyestrone, also showed a statistically significant improvement.
The results of this pilot trial suggests that this combination isoflavone nutritional supplement may significantly relieve the most troubling symptoms of menopause, as well as confer some chemopreventive and cardioprotective benefits.

ABSTRACT: The potential of Trifolium pratense (red clover) extract in the prevention of lipid disorder has attracted increasing attention in recent years. In this study, the aim was to determine whether and how red clover extract affected the development of murine diet-induced non-alcoholic steatohepatitis. Non-alcoholic steatohepatitis was induced in C57BL/6 mice by feeding mice with a methionine-choline-deficient (MCD) diet. Hematoxylin and eosin staining was used for histological analyses. Real-time PCR was used to analyze the mRNA expression levels. Hepatic steatosis and necroinflammation was observed in MCD diet-fed mice, and this diet-induced steatosis was significantly attenuated, whereas liver inflammation was not significantly attenuated, by red clover extract treatment. Consistent with the results of H&E staining, the MCD diet-induced increase of liver triglycerides and cholesterol levels were significantly reduced by red clover extract treatment. However, with the improvement in hepatic steatosis, mRNA levels of acetyl CoA oxidase, carnitine palmitoyl transferase-1, and liver fatty acid-binding protein, three genes regulated by peroxisome proliferator-activated receptor (PPAR) α, were unaffected. ABSTRACT: To investigate the effects of red clover extract on the blood glucose and lipid levels of type 2 diabetic db/db mice, male db/db mice were treated with this extract for a period of 5 weeks. The red clover extract had a significant effect on lowering the blood glucose levels of db/db mice. The serum triglyceride, serum total cholesterol, liver triglyceride and liver cholesterol levels for diabetic mice receiving red clover extract were significantly lower compared to those of the untreated diabetic mice. The mRNA expression of two target genes transcriptionally regulated by peroxisome proliferator-activated receptor (PPAR)γ was determined by quantitative real-time RT-PCR, and red clover extract was observed to significantly upregulate hepatic glucokinase and CD36 expression. Four target genes transcriptionally regulated by PPARα were also assayed, and red clover extract was observed to significantly downregulate hepatic apolipoprotein C3 expression whereas it had no significant effect on apolipoprotein A5, acetyl CoA oxidase and carnitine palmitoyl transferase-1 expression. In addition, hepatic mRNA expression of fatty acid synthase was also observed to be downregulated by red clover extract treatment. Thus, we conclude that red clover extract significantly improves the glucose and lipid homeostasis in db/db diabetic mice and that these effects are achieved at least in part by activating hepatic PPARα/γ and by inhibiting hepatic fatty acid synthase.Experimental and therapeutic medicine 10/2012; 4(4):699-704.
