CD14 and CD169 expression in human lymph nodes and spleen: specific expansion of CD14(+)CD169(-) monocyte-derived cells in diffuse large B-cell lymphomas

ABSTRACT The mononuclear phagocyte system of human lymphoid tissue comprises macrophages and dendritic cells (DCs). The heterogeneity of the non-DC mononuclear phagocyte population in human lymphoid tissue has been little addressed. Here, we studied the expression of 2 monocyte-derived markers, CD14 and CD169 (sialoadhesin), in reactive human lymphoid tissue as well as in a series of 51 B-cell lymphomas by immunohistochemistry on paraffin-embedded tissue. We confirmed that lymph node sinusoidal monocyte-derived cells were the only population staining for CD169. Although most sinusoidal histiocytes also expressed CD14, monocyte-derived cells with phagocytosis such as erythrophagocytosis, anthracosis, or tingible bodies macrophage lacked CD14 and CD169. Among B-cell lymphomas, splenic marginal zone lymphoma was the only one associated with an expansion of the CD14(+)CD169(+) cells in the cords. With respect to nodal B-cell lymphomas, CD14(+) cells were rare among B-chronic lymphocytic leukemia, follicular lymphoma (FL), mantle cell lymphoma (MCL). However, strikingly, we found a strong expansion of CD14(+)CD169(-) cells in numerous diffuse large B-cell lymphomas (DLBCLs), except in cases associated with numerous mitoses, apoptotic bodies, and tingible bodies macrophages. When cultivated in granulocyte/macrophage colony stimulating factor/interleukin 4, DLBCL purified CD14(+) cells differentiate into plasmacytoid cells, expressing DC-specific intercellular adhesion molecule 3-grabbing nonintegrin, suggesting dendritic cell differentiation potential. Our observation fits well with the lymph node and host response cluster signatures described in the gene profiling signatures of DLBCL. However, the role of this CD14(+) population that may constitute a microenvironment-related marker of this subgroup of DLBCL remains to be determined.

    ABSTRACT: In this study we assessed the prognostic significance of absolute monocyte count and elected the best cut-off value at diagnosis, in a large cohort of patients with diffuse large B-cell lymphoma. Data were retrieved for therapy-naive patients with diffuse large B-cell lymphoma followed in Israel and Italy during1993-2010. A final cohort of 1017 patients was analyzed with a median follow up of 48 months and a 5 year overall survival rate of 68%. The best absolute monocyte count cut-off level was< 630/mm3and the 5 years overall survival for these patients was 71% and 59% for those with >630mm3 (p=0.0002). Of the 1017 patients, 521 (51%) were treated with chemo-immunotherapy, and in this cohort, using multivariate analysis, elevated monocyte counte retained a negative prognostic value even when adjusted for IPI (HR 1.54, P=0.009). This large study shows that a simple parameter like absolute monocyte count (>630 /mm3) can easily be used routinely in the evaluation of newly diagnosed diffuse large B-cell lymphoma to identify high-risk patients with a worse survival in the rituximab era.

    ABSTRACT: Malignant transformation of cells is associated with aberrant glycosylation presented on the cell-surface. Commonly observed changes in glycan structures during malignancy encompass aberrant expression and glycosylation of mucins; abnormal branching of N-glycans; and increased presence of sialic acid on proteins and glycolipids. Accumulating evidence supports the notion that the presence of certain glycan structures correlates with cancer progression by affecting tumor-cell invasiveness, ability to disseminate through the blood circulation and to metastasize in distant organs. During metastasis tumor-cell-derived glycans enable binding to cells in their microenvironment including endothelium and blood constituents through glycan-binding receptors - lectins. In this review, we will discuss current concepts how tumor-cell-derived glycans contribute to metastasis with the focus on three types of lectins: siglecs, galectins, and selectins. Siglecs are present on virtually all hematopoietic cells and usually negatively regulate immune responses. Galectins are mostly expressed by tumor cells and support tumor-cell survival. Selectins are vascular adhesion receptors that promote tumor-cell dissemination. All lectins facilitate interactions within the tumor microenvironment and thereby promote cancer progression. The identification of mechanisms how tumor glycans contribute to metastasis may help to improve diagnosis, prognosis, and aid to develop clinical strategies to prevent metastasis.

    ABSTRACT: Ultra wide-band (UWB) radio uses base-band pulses of very short duration, thereby spreading the energy of radio signal very thinly over gigahertz. Power spectral density (PSD) of UWB signals consists of continuous and discrete components. Since the discrete component contributes more to PSD than continuous component, it presents greater interference to other narrow-band wireless systems. UWB was approved by the FCC for commercial deployment in 2002. In order to ensure negligible mutual interference, the FCC has specified emission limits for UWB applications. UWB is now under consideration as an alternative physical layer technology for wireless PAN in IEEE 802.15.3a. However, little study has been reported on PSD issues in IEEE 802.15.3a. This paper presents analysis on PSD of UWB signals in IEEE 802.15.3a systems, and on what frame reversion can achieve in reducing the PSD.
