Synthesis of Docetaxel and Butitaxel Analogues through Kinetic Resolution of Racemic β-Lactams with 7- O -Triethylsilylbaccatin III

ABSTRACT The kinetic resolution of racemic cis-4-phenyl- and cis-4-tert-butyl-3-hydroxy-beta-lactam derivatives with 7-O-triethylsilylbaccatin III yielded paclitaxel and butitaxel analogues with high diastereoselectivity. The results demonstrated that the tert-butyldimethylsilyl protecting group at the C3-hydroxy group of the beta-lactams provided optimum kinetic resolution in comparison with the sterically less demanding triethylsilyl group and the larger triisopropylsilyl group. In addition, it was found that the C4 beta-lactam substituents also influenced diastereoselectivity. The C4 tert-butyl-beta-lactams provided better diastereoselectivity than the corresponding C4 phenyl beta-lactams.

    ABSTRACT: β-lactam antibiotics has had a central role in the fight against pathogenic bacteria. It is the most widely employed family of antimicrobial agents to date. Central to β-lactam is the 2-azetidinone nucleus and is significant as bioactive agent. β-lactams are also a versatile building block for the stereoselective synthesis of non-β-lactam products. The antibacterial properties of the β-lactam antibiotics arise from an acylation process in which the N1-C2 bond of the four-membered 2-azetidinone ring is opened. The construction of functionalized chiral acyclic and heterocyclic building blocks is made possible by the 2-azetidinones since it has a high chirality content that can be transferred into a variety of products. As such, the stereoselective synthesis of different sized heterocycles of biological significance has been accomplished. The synthesis of different-sized heterocycles can be done through three through six-membered rings. Medium sized rings can be employed by employing the cyclic 2-azetidinone skeleton as a template on which to build the medium-sized ring using the chirality and functionalization of the β-lactam nucleus as a stereo-controlling element. Synthesis of macrolide core of cryptophycins can be accomplished by β-lactam and for macrocycles. The importance of β-lactams as synthetic intermediates has been widely recognized in organic synthesis because ring cleavage of any of the four single bonds of the β-lactam system is enhanced by ring strain.

    ABSTRACT: Quinoline analog 2-aryl-4H-3,1-benzothiazine derivatives 8–13, obtained by the condensation of o-aminobenzyl chloride 1 with substituted thiobenzamides 2–7, were transformed to azeto[2,1-a][3,1]benzothiazin-1-one derivatives 18–23a,b,c and 24d,e by reaction with the corresponding substituted acetyl chlorides 14–17 in the presence of triethylamine. The structures of the new molecules were determined by NMR spectroscopy and electron ionization (EI) mass spectrometry. The typical EI+ mass spectrometric fragmentations of 8–13 and 18–23a,b,c and 24d,e are discussed in detail.Graphical abstractA series of novel monochloro-, dichloro-, and phenyl-substituted (R3 or/and R4) β-lactams condensed with 3,1-benzothiazines were obtained by Staudinger reaction and studied by NMR spectroscopy and mass spectrometry.
