SPR biosensor for the detection of L-monocytogenes using phage-displayed antibody

ABSTRACT Whole cells of Listeria monocytogenes were detected with a compact, surface plasmon resonance (SPR) sensor using a phage-displayed scFv antibody to the virulence factor actin polymerization protein (ActA) for biorecognition. Phage Lm P4:A8, expressing the scFv antibody fused to the pIII surface protein was immobilized to the sensor surface through physical adsorption. A locally constructed fluidics system was used to deliver solutions to the compact, two-channel SPREETA sensor. Specificity of the sensor was tested using common food-borne bacteria and a control phage, M13K07 lacking the scFv fusion on its coat protein. The detection limit for L. monocytogenes whole cells was estimated to be 2 x 10(6)cfu/ml. The sensor was also used to determine the dissociation constant (Kd) for the interaction of phage-displayed scFv and soluble ActA in solution as 4.5 nM.

    ABSTRACT: Surface plasmon resonance (SPR) sensor, an optical sensor exploiting special electromagnetic waves-surface plasmon polaritons to probe interactions between an analyte in solution and a molecular recognition element immobilized on the SPR sensor surface, has been widely used in various realms, such as investigating bio-molecular interactions and binding properties, detection of biological and chemical analytes, environmental monitoring, food safety and medical diagnostics. This paper reviews the development of SPR sensors and SPR commercial instruments, and emphatically introduces the time-resolved surface plasmon resonance (TR-SPR) techniques. The excellent performances of high sensitivity and rapid detection are easily achieved with TR-SPR spectrometer, whereas the traditional SPR spectrometer cannot be accomplished. Therefore, TR-SPR spectrometer is appropriate for real time analysis of bio-recognition events and small molecular dy-namics. However, only two commercial TR-SPR instruments have been exhibited at present, Thermo FT-SPR 100 and Autolab ESPRIT (SPRINGLE). Unfortunately, the high-priced instruments are not suitable for extensive applications in laboratories at present. Herein, a novel commercial TR-SPR spectrometer has been introduced in this review.

    ABSTRACT: We identified 1219 articles published in 2006 that described work performed using commercial optical biosensor platforms. It is interesting to witness how the biosensor market is maturing with an increased number of instrument manufacturers offering a wider variety of platforms. However, it is clear from a review of the results presented that the advances in technology are outpacing the skill level of the average biosensor user. While we can track a gradual improvement in the quality of the published work, we clearly have a long way to go before we capitalize on the full potential of biosensor technology. To illustrate what is right with the biosensor literature, we highlight the work of 10 groups who have their eye on the ball. To help out the rest of us who have the lights on but nobody home, we use the literature to address common myths about biosensor technology.

    ABSTRACT: The incidence of foodborne infectious diseases is stable or has even increased in many countries. Consequently, our awareness regarding hygiene measures in food production has also increased dramatically over the last decades. However, even today's modern production techniques and intensive food-monitoring programs have not been able to effectively control the problem. At the same time, increased production volumes are distributed to more consumers, and if contaminated, potentially cause mass epidemics. Accordingly, research directed to improve food safety has also been taken forward, also exploring novel methods and technologies. Such an approach is represented by the use of bacteriophage for specific killing of unwanted bacteria. The extreme specificity of phages renders them ideal candidates for applications designed to increase food safety during the production process. Phages are the natural enemies of bacteria, and can be used for biocontrol of bacteria without interfering with the natural microflora or the cultures in fermented products. Moreover, phages or phage-derived proteins can also be used to detect the presence of unwanted pathogens in food or the production environments, which allows quick and specific identification of viable cells. This review intends to briefly summarize and explain the principles and current standing of these approaches.
