Human 17beta-hydroxysteroid dehydrogenase type 1 (17beta-HSD1) catalyzes the reduction of the weak estrogen estrone (E1) to the highly potent estradiol (E2). This reaction takes place in the target cell where the estrogenic effect is exerted via the estrogen receptor (ER). Estrogens, especially E2, are known to stimulate the proliferation of hormone-dependent diseases. 17beta-HSD1 is overexpressed in many breast tumors. Thus, it is an attractive target for the treatment of these diseases. Ligand- and structure-based drug design led to the discovery of novel, selective, and potent inhibitors of 17beta-HSD1. Phenyl-substituted bicyclic moieties were synthesized as mimics of the steroidal substrate. Computational methods were used to obtain insight into their interactions with the protein. Compound 5 turned out to be a highly potent inhibitor of 17beta-HSD1 showing good selectivity (17beta-HSD2, ERalpha and beta), medium cell permeation, reasonable metabolic stability (rat hepatic microsomes), and little inhibition of hepatic CYP enzymes.

    ABSTRACT: 17beta-Hydroxysteroid dehydrogenase type 1 (17beta-HSD1) catalyses the intracellular conversion of oestrone (E1) to oestradiol (E2). E2 is known to be involved in the development and progression of breast cancer and endometriosis. Since 17beta-HSD1 is overexpressed in these oestrogen-dependent diseases, inhibition of this enzyme may be a more target-directed therapeutical approach compared to established medical treatments. For the identification of highly active and selective 17beta-HSD1-inhibitors that are suitable for application as potential therapeutics, there is a need for an appropriate, efficient and reliable screening system. Here, we report the development and application of our screening system using our in house library of potential 17beta-HSD1-inhibitors. Four potent and selective compounds with a good first pharmacokinetic profile were identified.

    ABSTRACT: In order to become and remain competitive, organizations across America are moving away from traditional hierarchical management toward a network of small work teams. Recent surveys suggest that as many as 80% of American businesses now have one or more work teams in place. Such teams, dedicated to the continuous improvement of their piece of the business, have the potential of becoming the primary unit of performance in these new corporate systems. Many self-directed, problem solving and cross-functional teams are producing dramatic results in quality, productivity and speed. Far too many, however, are failing to realize their potential. Some never seem to get off the ground while others plateau or even drop off in performance after a promising start. For many organizations, keeping the momentum going is even more of a challenge than getting them started in the first place. Based on the author's experience in training and developing high performance teams, this paper identifies the three major factors that contribute to team performance. Additionally, it describes fifteen characteristics that must be addressed in order for work teams to reach their performance potential. Using these characteristics as a checklist can be helpful when establishing new work teams. But they may be even more useful when attempting to determine why existing work teams have leveled off or even decreased in performance

    ABSTRACT: This paper reports on the preliminary development of an improved defense technology and industrial base (DTIB) forecasting methodology. The approach examined several noted assessment and quality programs, began with previous industrial base assessment methods, and incorporated current quality perspectives. The methodology augments previous methods by improving the metrics of technology, integrated design, production processes, quality, and modern management and operations methods such as those of lean manufacturing. The resultant DTIB representation was reviewed by government and industry experts. Conclusions are presented. A preliminary set of measurement criteria was developed and sources for the data were established. A larger measurement set is needed to test the validity of the characteristics to represent the DTIB in the forecasting task
