Disulfide-containing cross-linked PEI derivative synthesized by click chemistry for non-viral gene delivery.

ABSTRACT Novel reducible disulfide-containing cross-linked polyethylenimines (PEI-SS-CLs) with high molecular weight (M(W)) were synthesized by click chemistry between disulfide-containing dialkyne cross-linkers and the azide functionalized low M(W) PEIs and evaluated as gene carriers. In vitro experiments revealed that the reducible PEI-SS-CLs were less cytotoxic and more effective in the gene transfection (both in the presence and absence of serum) than branched PEI (25kDa). These polymers are therefore attractive candidates for further in vivo evaluations.
