Synthesis and cytotoxicity of O,O '-dialkyl {[2-(substituted phenoxy) acetamido](substituted phenyl)methyl}phosphonates

ABSTRACT A series of O,O'-dialkyl {[2-(substituted phenoxy)acetamido](substituted phenyl)methyl}phosphonates was synthesized and their cytotoxic activities were tested against various human tumor cell lines. Some compounds (5q, 5r, 5s, 5w, 5x and 5y) showed relatively high cytotoxicity. Especially, compounds 5x and 5q exhibited the best cytotoxicity against KB and CNE2 cells with IC(50) 7.1 and 11.4 μM, respectively. Their inhibitory activities against KB and CNE2 cells were even higher than that of fluorouracil.

    ABSTRACT: Here we report various rigid alkanediamide derivatives, with three different substitution patterns of phosphonate ester for investigation of organogelation properties. The gelation properties of these compounds can be tuned by altering the number and position of the phosphonate esters. It was found that supramolecular self-assembly of the bisphosphonate esters (1a, 1d, 1h and 2a–b) resulted gel formation, whereas that of the tetraphosphonate esters (3a, 3d, 3h) resulted in construction of nanospheres and microspheres. The diverse morphology of the gel formation appears to depend predominantly on the substituted phenyl ring of the phosphonate ester functionality. Subsequent in vitro cytotoxic screening against four human tumour cell lines (A549, MDA-MB-231, MCF-7 and HeLa) using MTT assays revealed that derivatives 1f–h and 3a show cytotoxic potencies at concentrations less than 5 μM (IC50 values). Compounds 1f–h exhibited promising activity against A549 cell line, 1g–h were highly effective against MDA-MB-231, 1f and 3a show potential against MCF-7 and 1f–g were active against the HeLa cell line.

    ABSTRACT: Twelve new salicyl acyloxy phosphonate derivatives were synthesized via microwave-assisted reactions of salicylic acid with alpha-hydroxyphosphonate in DCC/DMAP. The structures of the products were confirmed by elemental analysis, IR, H-1 NMR and C-13 NMR data. The suitable synthetic conditions were as follows: in 15 mL of CH2Cl2 with power 800 W at room temperature for 3.5 h. The cytotoxic activities of all compounds on A-549, SGC-7901, HEp-2 and EC-109 in vitro were evaluated by using an 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method, and some of them showed good antitumor activity. Among the active compounds, espectively, compound 2f had better inhibition effect on HEp-2 cells growth [IC50=(13.9 +/- 0.6) mu mol/L], the IC50 values of compound 2k was (8.5 +/- 0.9) mu mol/L against EC-109 cells, similar to cisplatin [IC50=(5.9 +/- 1.0) mu mol/L].

    ABSTRACT: A series of novel molecules 5a-g containing N-aryl maleimide and α-hydroxyphosphonate moieties were synthesized. A distinct approach for high-yielding synthesis of α-hydroxyphosphonates has been discovered using various catalyst and solvents. The structures of the synthesized compounds were elucidated by IR, NMR, MS and CHN analysis. All the synthesized compounds were tested for qualitative (Zone of inhibition) and quantitative (MIC) antimicrobial activities against two pathogenic bacteria such as Bacillus subtilis (NCIM 2250) and Escherichia coli (ATCC 25922) and four pathogenic fungi such as Candida albicans (MTCC 277), Candida tropicalis (MTCC184), Aspergillus niger (MCIM 545) and Aspergillus clavatus (MTCC 132). The investigation of antimicrobial screening data revealed that most of the tested compounds are moderate to good microbial inhibitors.
