ABSTRACT Synthetic biology has advanced the design of genetic devices that can be used to reprogram metabolic activities in mammalian cells. By functionally linking the signal transduction of melanopsin to the control circuit of the nuclear factor of activated T cells, we have designed a synthetic signaling cascade enabling light-inducible transgene expression in different cell lines grown in culture or bioreactors or implanted into mice. In animals harboring intraperitoneal hollow-fiber or subcutaneous implants containing light-inducible transgenic cells, the serum levels of the human glycoprotein secreted alkaline phosphatase could be remote-controlled with fiber optics or transdermally regulated through direct illumination. Light-controlled expression of the glucagon-like peptide 1 was able to attenuate glycemic excursions in type II diabetic mice. Synthetic light-pulse-transcription converters may have applications in therapeutics and protein expression technology.

    ABSTRACT: La biologie de synthèse est un domaine émergent en quête d’outils afin de formaliser et d’automatiser la caractérisation et la conception de systèmes biologiques. Dans ce cadre, nous proposons de définir le langage GUBS (Genetic Unified Behaviour Specification) utilisé pour spécifier de manière comportementale les systèmes biologiques à synthétiser. De cette définition, le compilateur Ggc recherche un assemblage de composants biologiques capable de reproduire le comportement spécifié. Gubs est un langage déclaratif que nous pouvons assimiler à un langage de requêtes évolué de recherche de comportements décrits par un ensemble de relations causales. Associé à ce langage, le compilateur Ggc sélectionne automatiquement les composants pour qu’une fois assemblés leur comportement collectif "inclut" celui décrit. La compilation d’une spécification Gubs s’appuie sur le principe d’ACI-Unification en utilisant plus généralement un schéma similaire au système de preuve automatique qui permet de garantir formellement la correction de cet assemblage.

    ABSTRACT: Mammals rely upon three ocular photoreceptors to sense light: rods, cones, and intrinsically photosensitive retinal ganglion cells (ipRGCs). Rods and cones resolve details in the visual scene. Conversely, ipRGCs integrate over time and space, primarily to support "non-image" vision. The integrative mechanisms of ipRGCs are enigmatic, particularly since these cells use a phototransduction motif that allows invertebrates like Drosophila to parse light with exceptional temporal resolution. Here, we provide evidence for a single mechanism that allows ipRGCs to integrate over both time and wavelength. Light distributes the visual pigment, melanopsin, across three states, two silent and one signaling. Photoequilibration among states maintains pigment availability for sustained signaling, stability of the signaling state permits minutes-long temporal summation, and modest spectral separation of the silent states promotes uniform activation across wavelengths. By broadening the tuning of ipRGCs in both temporal and chromatic domains, melanopsin tristability produces signal integration for physiology and behavior. Copyright © 2015 Elsevier Inc.     ABSTRACT: Nucleic acid vaccines have been gaining attention as an alternative to the standard attenuated pathogen or protein based vaccine. However, an unrealized advantage of using such DNA or RNA based vaccination modalities is the ability to program within these nucleic acids regulatory devices that would provide an immunologist with the power to control the production of antigens and adjuvants in a desirable manner by administering small molecule drugs as chemical triggers. Advances in synthetic biology have resulted in the creation of highly predictable and modular genetic parts and devices that can be composed into synthetic gene circuits with complex behaviors. With the recent advent of modified RNA gene delivery methods and developments in the RNA replicon platform, we foresee a future in which mammalian synthetic biologists will create genetic circuits encoded exclusively on RNA. Here, we review the current repertoire of devices used in RNA synthetic biology and propose how programmable 'smart vaccines' will revolutionize the field of RNA vaccination.
