ABSTRACT Treatment of nitroso Diels-Alder cycloadducts 1 with diazomethane in the presence of palladium acetate gives synthetically useful exo-6-oxa-7-azatricyclo[,4]octane derivatives 7 in good to excellent yield. Using this methodology, a conformationally restricted 2',3'-methano carbocyclic nucleoside was efficiently synthesized from nitroso cycloadduct 1a in 7 steps.

    ABSTRACT: Several novel oxazolidinone antibiotics with a spiropiperazinyl substituent at the 4'-position of the phenyl ring were synthesized through nitroso Diels-Alder chemistry and the in vitro antibacterial activities were evaluated against various Gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus, Enterococcus faecalis), Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa) and mycobacteria (Mycobacterium vaccae, Mycobacterium tuberculosis). Analogs (8a and 12) were active against selected drug resistant microbes, like methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE) and had no mammalian toxicity in a Hep-2 cellular assay (CC(50) >100 μM).

    ABSTRACT: Cyclopropane is often present in natural and biologically active products. Alternatively, the cyclopropane structure has been used as parts for the modification of such products. It has a high ring strain because of its bond angle, and this property facilitates unique reactions. This chapter collects recent representative examples of [2+1]-type cyclopropanation reactions. Specifically, it reviews six categories of reactions: Michael-induced ring closure (MIRC) reaction, the Simmons-Smith reaction, reactions by carbenes from diazo-alkanes catalyzed/noncatalyzed by transition metals, cyclo-isomerization reactions by transition metal catalysts, the Kulinkovich reaction, and miscellaneous reactions. The asymmetric synthesis of cyclopropanes, which is a topic of interest among synthetic chemists, is discussed in each category.

    ABSTRACT: Our flow reaction systems have provided quantitative yields of nitroso Diels-Alder products with no byproducts in cases of cyclic dienes without temperature and pressure controls. Additionally, the reaction times were significantly shortened by using homogeneous catalyst (CuCl) or heterogeneous reagent (MnO2) in comparison with batch reaction.
