Pengaruh Suhu Dan Lama Penyimpanan Pada kestabilan Sampel Enzim Asetilkolonesterase Dari Darah Manusia

ABSTRACT 09E02229 Penelitian tentang Pengaruh Suhu Dan Lama Penyimpanan Pada Kestabilan Sampel Enzim Asetilkolinesterase Dari Darah Manusia dilaksanakan pada bulan Maret hingga bulan Mei 2008 di laboratorium Terpadu Universitas Sumatera Utara. Pengambilan sampel pada penelitian ini dilakukan dengan teknik acak sederhana (simple random sampling) dari darah 10 pria sehat usia 18-25 tahun (mahasiswa Universitas Tjut Nyak Dhien Medan). Rancangan yang digunakan pada penelitian ini adalah Rancangan Acak Lengkap (RAL) Faktorial, dan analisa statistik dilakukan dengan ANOVA. Plasma darah yang diperoleh diukur aktifitas enzim asetilkolinesterase (0 minggu), lalu plasma darah tersebut disimpan pada suhu -20ºC, 4-6ºC, dan 25-30ºC selama 2 dan 4 minggu kemudian dilakukan pengukuran kembali aktifitas enzim asetilkolinesterase. Pengukuran aktifitas enzim asetilkolinesterase dilakukan dengan menggunakan alat spektrofotometer (genesys 5). Hasil yang diperoleh dari penelitian ini menunjukkan bahwa plasma darah yang disimpan pada suhu -20ºC dan 4-6ºC selama 4 minggu tidak mempengaruhi terhadap kestabilan dan aktifitas enzim asetilkolinestrase. Sedangkan penyimpanan pada suhu 25-30ºC selama 2 - 4 minggu berpengaruh terhadap kestabilan aktifitas enzim tersebut. The research about Effect on Temperature And the Storage length of Stability of Sample Enzyme Achetylcolinesterase From Blood Human which was conducted on March until May 2008 at the Interrelated Discipline Laboratory of University of North Sumatra. The sample of this research has been taken by way of complete simple random sampling from 10 healthy male and with the age of 18 up to 25 (University of Tjut Nyak Dhien Medan). The method used in this research is complete factorial random, and statistic analysis conducted by way of variant analysis (ANOVA). The obtained Blood plasma is measured through the activities of enzyme achetylcolinesterase (zero week), then the blood plasma was stored in the temperature of -20ºC, 4-6ºC, and 25-30ºC within 2 and 4 weeks then the re-measurement is conducted on the activity of Enzyme Achetylcolinesterase. In term of the measurement of enzyme achetylcolinesterase activity, spectrophotometer (genesys 5). The result of this research, the blood plasma which was kept in the temperature of -20ºC and 4-6ºC did not affect the stability of enzyme achetylcolinesterase activities within 4 weeks, while the blood plasma that was stored in the temperature of 25-30ºC and the enzyme achetylcolinesterase activity tends to change after being stored within 2- 4 weeks. dr.
