Synthesis and antimicrobial activity of some novel ciprofloxacin analogues

From the cyclisation of potassium dithiocarbazinate with hydrazine hydrate in water, 4-(amino)-5-phenyl - 4H-1, 2, 4-triazole- 3- thiol belonging to the 1,2,4- triazole derivative have been prepared. The different aromatic aldehydes such as benzaldehyde, 2-hydroxy benzaldehyde, 2,4-dichloro benzaldehyde, 3-methoxy 4-hydroxy benzaldehyde, 4-dimethyl amino benzaldehyde and furfuraldehyde have been attached to the triazole through the Schiff reaction to give different Schiff bases. Ciprofloxacin have been incorporated to the new series of Schiff bases of 1,2,4- triazole via Mannich reaction. The new compounds have been evaluated in vitro for their antimicrobial activity against B.subtilis, K.Pneumoniae, and P.aeruginosa. All the compounds showed in vitro gram positive and gram negative activity generally comparable or superior to that of reference ciprofloxacin.

    ABSTRACT: Five NH-derivatives of ciprofloxacin (2-6) have been prepared by acetylation/benzoylation on ciprofloxacin (1) in order to carry out screening for antibacterial activities on some gram-positive and gram-negative bacteria. The antifungal activity of these compounds has also been made against Candida albicans. All of the derivatives have shown enhanced activities against gram-negatives bacteria than parent antibiotic, ciprofloxacin among which compounds 2 and 6 are the most potent agents. Regarding the antifungal activity all of the compounds have shown highest activity than ciprofloxacin. All the compounds have been characterized with spectral analysis.

    ABSTRACT: The carboxylic acid group of ciprofloxacin was modified and amino mercapto triazole was substituted. The compound was confirmed by physical parameters (solubility, melting point), chromatographic methods (TLC) and consistent with its IR & 1HNMR spectra. The synthesized analogue was screened for antibacterial activity against one gram positive & two gram negative species. The compound exhibited good antibacterial effect towards gram negative species when compared to the standard ciprofloxacin. At the same time the analogue was retaining antibacterial activity towards gram positive species when compared to standard ciprofloxacin. The molecular docking studies showed a good correlation between their antibacterial activity and autodock binding free energy.

    ABSTRACT: A number of novel ofloxacin analogues were synthesized by modifying the carboxylic acid at C-6. To investigate the antimicrobial data on structural basis, in-silico docking studies of the tested compounds into the crystal structure of topoisomerase II using Autodock vina 4.0 program was performed in order to predict the affinity and orientation of the synthesized compounds at the activities. R 2 values show good agreement with predicted binding affinities obtained by molecular docking studies. Also, it is verified by in-vitro antimicrobial screening, where all the compounds were most active against Staphylococcus aureus, Staphylococcus epidermidis and Bacillus subtilis. Among these compounds 3a, 3b, 3f showed good MIC (0.125 μg/ml).
