Acondicionamiento osmótico de semillas de tomate de cáscara

Con el objetivo de evaluar el efecto del acondicionamiento osmótico sobre la calidad fi siológica de semillas de tomate de cáscara, se realizó un experimento en el Laboratorio de Análisis de Semillas del Colegio de Postgraduados en Montecillo, Estado de México, México, en el período septiembre de 2004 a mayo de 2005. La semilla de tomate de cáscara cv. Rendidora fue osmoacondicionada con KNO3, KCl y polietilen glicol-8000 a -5, -10, -15 y -20 atm durante 48, 72 y 96 h y posteriormente se almacenó durante 0, 7, 90 y 180 días. Los parámetros evaluados fueron: porciento de germinación, plántulas anormales, semillas latentes y semillas muertas, tiempo para alcanzar 50% de germinación y peso seco de plántula. El efecto del acondicionamiento osmótico sobre las variables de germinación varió en función del agente osmótico y su potencial, y la duración del tratamiento. El almacenamiento favoreció el porciento de germinación y redujo el de semillas latentes. El acondicionamiento osmótico con KCl a -15 atm durante 48 h o con KNO3 a -20 atm durante 48 h, mejoraron la calidad fi siológica de la semilla de tomate de cáscara

    ABSTRACT: Priming seeds of spinach (Spinacia oleracea L.) by extended, partial imbibition in osmotic solutions of polyethylene glycol “6000” (PEG) markedly improved subsequent germination at 30°C. The optimum conditions for priming were −12.5 bars water potential in the osmoticum when seeds were imbibed for 14 days at 10°C. Primed seeds could be dried back and stored for at least 30 days at 5°C without losing their improved capacity for germination at high temperature.

    ABSTRACT: Abscisic acid-pretreated carrot (Daucus carota) somatic embryos survive dehydration upon slow drying, but fast drying leads to poor survival of the embryos. To determine whether the acquisition of desiccation tolerance is associated with changes in the physical stability of the cytoplasm, in situ Fourier transform infrared microspectroscopy was used. Although protein denaturation temperatures were similar in the embryos after slow or fast drying, the extent of the denaturation was greater after fast drying. Slowly dried embryos are in a glassy state at room temperature, and no clearly defined glassy matrix was observed in the rapidly dried embryos. At room temperature the average strength of hydrogen bonding was much weaker in the rapidly dried than in the slowly dried embryos. We interpreted the molecular packing to be "less tight" in the rapidly dried embryos. Whereas sucrose (Suc) is the major soluble carbohydrate after fast drying, upon slow drying the trisaccharide umbelliferose accumulates at the expense of Suc. The possibly protective role of umbelliferose was tested on protein and phospholipid model systems, using Suc as a reference. Both umbelliferose and Suc form a stable glass with drying: They depress the transition temperature of dry liposomal membranes equally well, they both prevent leakage from dry liposomes after rehydration, and they protect a polypeptide that is desiccation sensitive. The similar protection properties in model systems and the apparent interchangeability of both sugars in viable, dry somatic embryos suggest no special role of umbelliferose in the improved physical stability of the slowly dried embryos. Also, during slow drying LEA (late-embryogenesis abundant) transcripts are expressed. We suggest that LEA proteins embedded in the glassy matrix confer stability to these slowly dried embryos.

    ABSTRACT: A protein named QP47 has been purified from quiescent nuclei of root meristems of Pisum sativum, and used to prepare a polyclonal antibody. Immunolocalization of this protein with fluorescent probes revealed a nuclear distribution of thread-like structures. However, the relationship between the distribution of QP47 immunofluorescence and the structural organization of the chromatin required further investigation. The decrease in content of this protein in the nuclei of embryo cells seems to be correlated with the transition from quiescence to proliferation. QP47 degradation seems to depend upon an increase in the state of its phosphorylation. This protein is not present in normally proliferating cells, or in cells whose cell proliferation has been arrested by starvation or differentiation. It is hypothesized that QP47 may be required specifically during the quiescent period for specific structural organization of the chromatin.
