Caracterización parcial del colágeno a partir del manto, aleta y tentáculos de calamar gigante (Dosidicus gigas)

ABSTRACT El colágeno en el Dosidicus gigas o calamar gigante participa en los mecanismos natatorios del organismo. Sin embargo, la información fisicoquímica sobre esta proteína es escasa. En este estudio se comparó el contenido y algunas propiedades fisicoquímicas de colágeno soluble en pepsina (CSP) e insoluble (CI) extraídos de manto, aleta y tentáculos de Dosidicus gigas fresco. El contenido de colágeno total, así como el perfil electroforético fueron diferentes para cada región anatómica evaluada. Los tentáculos presentaron el mayor contenido de CI. En estudios de microscopia electrónica de barrido (MEB) se observó un arreglo estructural similar en el CSP de las tres regiones y en el CI, marcadas diferencias en el empaquetamiento de las fibras. Las tres fracciones proteicas de colágeno, mostraron una alta resistencia térmica (115-120 °C), siendo superior para tentáculos en comparación con aleta y manto. Esta información contribuye a un mayor conocimiento de la especie y una mejor aplicación comercial de la misma.

    ABSTRACT: The effect of frozen storage (-10 and -30 degrees C), formaldehyde, and fish oil on collagen, isolated from cod muscle, was investigated. Salt- and acid-soluble collagen fractions as well as insoluble collagen indicated changes in solubility on frozen storage. Differential scanning calorimetry (DSC) showed a highly cooperative transition at 28.2 degrees C for isolated collagen. Changes in the thermodynamic properties of collagen were observed on frozen storage at -10 degrees C compared with the control at -30 degrees C because of changes in structure. In the presence of formaldehyde, there were no changes in the DSC collagen transition; however, in the presence of fish oil there was an increase in enthalpy and an extra peak was observed at 44.6 degrees C, indicating collagen-fish oil interaction. Structural changes resulted in a decrease in the solubility of collagen in salt and acid solution. FT-Raman spectra obtained for collagen at -10 degrees C and -30 degrees C confirmed the alteration of the conformation of collagen not only at -10 degrees C but also in the presence of formaldehyde and fish oil.

    ABSTRACT: Pyridinoline (Pyr), one of the mature crosslinks of collagen, was determined in muscular collagen of three species of fish. The amounts of muscular Pyr in red sea bream, yellowtail, and tiger puffer were 3.4, 8.8, and 50.3 mmol/mol collagen, respectively, indicating that the Pyr concentration in muscular collagen differs greatly among fish species. The Pyr concentration in tiger puffer muscular collagen was the greatest, but it was only one-fourth that in rabbit muscle. As in mammalian skin collagen, Pyr was not detected in skin collagen of red sea bream and yellowtail. However, tiger puffer skin contained Pyr (3.75 mmol/mol collagen). The presence of Pyr would have a relationship to some features of tiger puffer skin, such as mechanical strength and thickness. Pyr concentrations in acid-soluble collagen (ASC), pepsin-solubilized collagen (PSC), and insoluble collagen (ISC) in muscles of three species of fish were determined. Pyr was found in ISC > PSC > ASC, from the highest to the lowest concentration, and the concentration in ISC was 45–200 times that in ASC. Therefore, Pyr crosslinks that are formed between collagen molecules would have a close relationship to collagen solubility.
