Biodegradation Aspects of Polycyclic Aromatic Hydrocarbons (PAHs): A Review

PAHs are aromatic hydrocarbons with two or more fused benzene rings with natural as well as anthropogenic sources. They are widely distributed environmental contaminants that have detrimental biological effects, toxicity, mutagenecity and carcinogenicity. Due to their ubiquitous occurrence, recalcitrance, bioaccumulation potential and carcinogenic activity, the PAHs have gathered significant environmental concern. Although PAH may undergo adsorption, volatilization, photolysis, and chemical degradation, microbial degradation is the major degradation process. PAH degradation depends on the environmental conditions, number and type of the microorganisms, nature and chemical structure of the chemical compound being degraded. They are biodegraded/biotransformed into less complex metabolites, and through mineralization into inorganic minerals, H(2)O, CO(2) (aerobic) or CH(4) (anaerobic) and rate of biodegradation depends on pH, temperature, oxygen, microbial population, degree of acclimation, accessibility of nutrients, chemical structure of the compound, cellular transport properties, and chemical partitioning in growth medium. A number of bacterial species are known to degrade PAHs and most of them are isolated from contaminated soil or sediments. Pseudomonas aeruginosa, Pseudomons fluoresens, Mycobacterium spp., Haemophilus spp., Rhodococcus spp., Paenibacillus spp. are some of the commonly studied PAH-degrading bacteria. Lignolytic fungi too have the property of PAH degradation. Phanerochaete chrysosporium, Bjerkandera adusta, and Pleurotus ostreatus are the common PAH-degrading fungi. Enzymes involved in the degradation of PAHs are oxygenase, dehydrogenase and lignolytic enzymes. Fungal lignolytic enzymes are lignin peroxidase, laccase, and manganese peroxidase. They are extracellular and catalyze radical formation by oxidation to destabilize bonds in a molecule. The biodegradation of PAHs has been observed under both aerobic and anaerobic conditions and the rate can be enhanced by physical/chemical pretreatment of contaminated soil. Addition of biosurfactant-producing bacteria and light oils can increase the bioavailability of PAHs and metabolic potential of the bacterial community. The supplementation of contaminated soils with compost materials can also enhance biodegradation without long-term accumulation of extractable polar and more available intermediates. Wetlands, too, have found an application in PAH removal from wastewater. The intensive biological activities in such an ecosystem lead to a high rate of autotrophic and heterotrophic processes. Aquatic weeds Typha spp. and Scirpus lacustris have been used in horizontal-vertical macrophyte based wetlands to treat PAHs. An integrated approach of physical, chemical, and biological degradation may be adopted to get synergistically enhanced removal rates and to treat/remediate the contaminated sites in an ecologically favorable process.

    ABSTRACT: Benzo(a)pyrene (BaP) and dichlorodiphenyltrichloroethane (DDT) are persistent organic pollutants and environmental estrogens (EEs) with known toxicity towards the green mussel, Perna viridis. In this study, the toxic effects of BaP (10 μg/L) and DDT (10 μg/L) and their mixture were assessed in green mussel gills with proteomic and metabolomic approaches. Metabolic responses indicated that BaP mainly caused disturbance in osmotic regulation by significantly decrease in branched chain amino acids, dimethylamine and dimethylglycine in gills of male green mussels after exposure for 7 days. DDT mainly caused disturbance in osmotic regulation and energy metabolism by differential alteration of betaine, dimethylamine, dimethylglycine, amino acids, and succinate in gills of male green mussels. However, the mixture of BaP and DDT didn't show obvious metabolite changes. Proteomic analysis showed different protein expression profiles between different treatment groups, which demonstrated that BaP, DDT and their mixture may have different modes of action. Proteomic responses revealed that BaP induced cell apoptosis, disturbance in protein digestion and energy metabolism in gills of green mussels, whereas DDT exposure altered proteins that were associated with oxidative stress, cytoskeleton and cell structure, protein digestion and energy metabolism. However, the mixture of BaP and DDT affected proteins related to the oxidative stress, cytoskeleton and cell structure, protein biosynthesis and modification, energy metabolism, growth and apoptosis.

    ABSTRACT: We investigated the bioremediation of 16 polycyclic aromatic hydrocarbons (PAH) in historically creosote contaminated soil using both laboratory and field scale experiments. We found that nutrient amendments and circulation of methyl-β-cyclodextrin (CD) solution enhanced soil microbial degradation capacity. In the laboratory experiment, the degradation of lower molecular weight, 2-3 ringed PAHs was achieved already by circulating nutrient solution and the use of CD mainly increased the desorption and removal of larger, 4-5 aromatic ringed PAH compounds. The 1% CD concentration was most feasible for bioremediation as most of the extracted PAH compounds were degraded. In the 5% CD treatment, the PAH desorption from soil was too fast compared to the degradation capacity and 25% of the total PAH amount remained in the circulated solution. Similar lab-scale results have been generated earlier, but very little has been done in full field scale, and none in freezing conditions. Although freezing stopped circulation and degradation completely during the winter, PAH degradation returned during the warm period in the field test. Circulation effectiveness was lower than in the laboratory but the improved nutrient and moisture content seemed to be the main reason for decreasing soil PAH concentrations. It also appeared that PAH extraction yield in chemical analysis was increased by the CD treatment in field conditions and the results of CD-treated and non-treated soil may therefore not be directly comparable. Therefore, a positive effect of CD on PAH degradation velocity could not be statistically confirmed in the field test. Based on our results, we recommend initiating the bioremediation of PAH contaminated soil by enhancing the microbial degradation with nutrient amendments. The CD seems to be useful only at the later stage when it increases the solubilisation of strongly absorbed contaminants to some extent. More investigation is also needed of the CD effect on the PAH yield in the chemical analysis.

    ABSTRACT: Abstract Bioremediation is an effective strategy for cleaning up organic contaminants, such as polycyclic aromatic hydrocarbons (PAHs) and volatile organic compounds (VOCs). Advanced bioremediation implies that biotic agents are more efficient in degrading the contaminants completely. Bioremediation by microbial degradation is often employed and to make this process efficient, natural and cost-effective materials can serve as supportive matrices. Clay/modified clay minerals are effective adsorbents of PAHs/VOCs, and readily available substrate and habitat for microorganisms in the natural soil and sediment. However, the mechanism underpinning clay-mediated biodegradation of organic compounds is often unclear, and this requires critical investigation. This review describes the role of clay/modified clay minerals in hydrocarbon bioremediation through interaction with microbial agents in specific scenarios. The vision is on a faster, more efficient and cost-effective bioremediation technique using clay-based products. This review also proposes future research directions in the field of clay modulated microbial degradation of hydrocarbons.
