Risk of Non-Hodgkin Lymphoma Associated with Germline Variation in Genes that Regulate the Cell Cycle, Apoptosis, and Lymphocyte Development

ABSTRACT Chromosomal translocations are the hallmark genetic aberration in non-Hodgkin lymphoma (NHL), with specific translocations often selectively associated with specific NHL subtypes. Because many NHL-associated translocations involve cell cycle, apoptosis, and lymphocyte development regulatory genes, we evaluated NHL risk associated with common genetic variation in 20 candidate genes in these pathways. Genotyping of 203 tag single nucleotide polymorphisms (SNP) was conducted in 1,946 NHL cases and 1,808 controls pooled from 3 independent population-based case-control studies. We used logistic regression to compute odds ratios (OR) and 95% confidence intervals (CI) for NHL and four major NHL subtypes in relation to tag SNP genotypes and haplotypes. We observed the most striking associations for tag SNPs in the proapoptotic gene BCL2L11 (BIM) and BCL7A, which is involved in a rare NHL-associated translocation. Variants in BCL2L11 were strongly related to follicular lymphoma only, particularly rs3789068 (OR(AG), 1.41; 95% CI, 1.10-1.81; OR(GG), 1.65; 95% CI, 1.25-2.19; P(trend) = 0.0004). Variants in BCL7A were strongly related to diffuse large B-cell lymphoma only, particularly rs1880030 (OR(AG), 1.34; 95% CI, 1.08-1.68; OR(AA), 1.60; 95% CI, 1.22-2.08; P(trend) = 0.0004). The associations for both variants were similar in all three studies and supported by haplotype analyses. We also observed notable associations for variants in BCL6, CCND1, and MYC. Our results support the role of common genetic variation in cell cycle, apoptosis, and lymphocyte development regulatory genes in lymphomagenesis, and suggest that effects may vary by NHL subtype. Replication of our findings and further study to identify functional SNPs are warranted.

    ABSTRACT: Background: Chromosomal translocations are genetic aberrations associated with specific non-Hodgkin lymphoma (NHL) subtypes. This study investigated the differential gene expression profile of Egyptian NHL cases based on a microarray approach. Materials and Methods: The study included tissue samples from 40 NHL patients and 20 normal lymph nodes used as controls. Total RNA was extracted and used for cDNA microarray assays. The quantitative real time polymerase chain reaction was used to identify the aberrantly expressed genes in cancer. Results: Significant associations of 8 up-regulated and 4 down-regulated genes with NHL were observed. Aberrant expression of a new group of genes not reported previously was apparent, including down-regulated NAG14 protein, 3 beta hydroxy-delta 5-c27 steroid oxi-reductase, oxi-glutarate dehydrogenase (lipo-amide), immunoglobulin lambda like polypeptide 3, protein kinase x linked, Hmt1, and caveolin 2 Tetra protein. The up-regulated genes were Rb binding protein 5, DKFZP586J1624 protein, protein kinase inhibitor gamma, zinc finger protein 3, choline ethanolamine phospho-transferase CEPT1, protein phosphatase, and histone deacetylase-3. Conclusions: This study revealed that new differentially expressed genes that may be markers for NHL patients and individuals who are at high risk for cancer development.

    ABSTRACT: Summary form only given, as follows. The digital communication industry requires RF amplifiers that provide constant gain at steadily increasing rated output power levels. One method to obtain linear operation in a traveling wave tube (TWT) is to operate the device well below its saturated power and to use a multi-stage depressed collector for energy recovery. This method, while effective, still has the disadvantage of operating at lower device efficiency than possible near saturation. One approach regularly used for extending the linear output power range of the device is to use an external linearizer circuit to condition the drive to the tube. While effective, the linearizer presents additional cost to the system while providing diminished effectiveness close to saturation. It is of continuing interest, therefore, to investigate and develop high power TWT designs with ultra-linear phase and amplitude transfer characteristics. The design of a linear TWT was approached on several fronts. First, simple Pierce theory was used to obtain a baseline design and to investigate many of the tradeoffs between design parameters and the device transfer characteristics. Next, Christine 1D, a 1-dimensional, large-signal, helix TWT code developed by the Naval Research Laboratory was used to optimize and verify the design. Finally, Christine 3D, a 2 1/2-dimensional, large-signal, TWT code will be used to model RF beam expansion and to calculate the spent beam distribution for collector optimization. The optimized design is scheduled to be built and tested in the Spring, 2002. Available test data will be presented, as will the design tradeoffs and analysis.
