Genetic analysis and pathogenicity of betanodavirus isolated from wild redspotted grouper Epinephelus akaara with clinical signs.

ABSTRACT Diseased wild redspotted grouper Epinephelus akaara were collected from Seto Inland Sea, Ehime Prefecture, in August 2002. Fish showed erratic swimming behavior and inflation of the swim bladder. The fish brains were positive for nodavirus in both RT-PCR and nested PCR. The sequence of the nested PCR product (177 nt) was closely related to that of a known betanodavirus, redspotted grouper nervous necrosis virus. When juvenile sevenband grouper E. septemfasciatus were challenged intravitreously with virus, abnormal swimming behavior and high mortality were observed. This is the first report on viral nervous necrosis in a wild population of redspotted grouper with clinical signs.

    ABSTRACT: Piscine nodaviruses (Betanodaviridae) are frequently reported from a variety of cultured and wild finfishes. These non-enveloped, single-stranded RNA virions cause viral encephalopathy and retinopathy (VER), also known as viral nervous necrosis (VNN) or fish encephalitis. Recently, nodavirus infections have posed serious problems for larval and juvenile cultured halibut Hippoglossus hippoglossus in Norway and Scotland. To date, no such viruses have been described from any cultured or wild pleuronectid in Atlantic Canada. Obviously, there exists a need to survey wild populations of pleuronectids to assess the risk of potential transfer of nodavirus from wild to caged fishes. This paper presents the results of monthly surveys (April 2000 to March 2001) of viruses from wild winter flounder Pleuronectes americanus collected from Passamaquoddy Bay, New Brunswick, Canada. Tissue samples from wild flounder were screened initially on commercial cell lines (EPC, SSN-1, SHK and CHSE-214) for any evidence of cytopathic effect (CPE). After confirmation of CPE, nodavirus identification was achieved using reverse transcription polymerase chain reaction (RT-PCR) analysis. We detected nodavirus from only 1 out of 440 flounder (0.23%) examined. This is the first report of piscine nodavirus isolated from wild winter flounder in Atlantic Canada, and although this prevalence may seem low, we discuss the implications of this finding for Canada's emerging halibut aquaculture industry.

  • Article: Experimental vertical transmission of nodavirus from broodfish to eggs and larvae of the sea bass, Dicentrarchus labrax (L.)

    ABSTRACT: Adult broodstock sea bass, Dicentrarchus labrax (L.), were intramuscularly inoculated with live Sb-Atl nodavirus strain and maintained at two different water temperatures (13 and 20 °C) until spawning. Each spawn was then incubated separately in UV treated water for 3 days and newly hatched larvae reared for 22 days at 13–15 °C. The inoculated broodfish developed a specific immune response following inoculation with the live virus and specific antibodies were detected in the sera of all the inoculated fish whatever the temperature used (13 and 20 °C). Specific nodavirus antigen was also detected by enzyme-linked immunosorbent assay (ELISA) and nested reverse transcription-polymerase chain reaction (RT-PCR) in the brain and the liver of these fish. Vertical transmission of the nodavirus disease was demonstrated following inoculation of the broodfish with the live virus strain, as typical lesions associated with the nodavirus antigen were detected by immunohistochemistry in the brain of larvae at days 14–22 following hatching. The eggs from seven inoculated females that spawned during the experiment were tested for nodavirus by antigen ELISA, RT-PCR and nested RT-PCR. Fertilized and non-fertilized eggs, as well as developing larvae, were found nodavirus positive using these techniques. However, the nested RT-PCR was the only technique that could detect the nodavirus in ovarian biopsies before spawning. Therefore, the selection of nodavirus-free sea bass broodstock cannot be achieved by ELISA or RT-PCR testing of ovarian biopsies. Alternative methods such as serum nodavirus antibody detection of broodstock and/or nested RT-PCR testing of eggs before spawning should be used.

    Article: Mass mortalities associated with viral nervous necrosis (VNN) disease in two species of hatchery‐reared grouper, Epinephelus fuscogutatus and Epinephelus akaara (Temminck & Schlegel)

    ABSTRACT: Mass mortalities of hatchery-reared juvenile groupers have occurred in southern Taiwan. The diseased fish swam in a darting, corkscrew fashion. Light microscopy revealed vacuolation in the brain tissue. Electron microscopy showed numerous non-enveloped, cytoplasmic viral particles (20–25 nm in diameter) in the brain cells, and many virions were enclosed in the membrane-bound organelles of the cells. Two structural proteins of the purified grouper virus, with molecular weights of 44 and 43 kDa, were revealed by SDS-PAGE. Moreover, the results of RT-PCR and nested PCR diagnosis using primers specific to the T2 and T4 target segments of striped jack nervous necrosis virus (SJNNV) RNA2 genes suggest that this virus is a fish nodavirus, and is designated as GNNV 9410 strain (grouper nervous necrosis virus strain 9410). This is the first case report of viral nervous necrosis among marine fish in Taiwan.
