[Further biochemical investigations on glycosaminoglycans extracted from Euglena gracilis]

Polyanionic glycans extracted from Euglena gracilis have been studied by biochemical, chromatographic and electrophoretic analysis. Our results show the presence of a fraction which precipitate with CPC and another one which not precipitate with CPC. The CPC precipitable material fractionated on CPC-Cellulose column shows the presence of 5 Glycosaminoglycans; the not CPC precipitable material contains uronic acid, galactose, sulfate, galactosamine and cannot be related to Keratan sulfate.

The effect of benzyl beta-D-xyloside on keratan sulphate synthesis in ox articular cartilage

Cartilage from adult bovine hock joints was incubated with [3H]galactose or [35S]sulphate in the presence of benzyl beta-D-xyloside. The decrease in radioisotope incorporation into keratan sulphate in the presence of 1.0mM benzyl beta-xyloside was directly related to a reduction in the average length of the keratan sulphate chains. This effect of beta-xyloside on keratan sulphate biosynthesis was markedly different from its effect on chondroitin sulphate biosynthesis.

Glycosaminoglycans (GAGS) synthesis and accumulation on the cell surface of skin fibroblasts in two patients with Werner's syndrome were measured by the incorporation of 3H-glucosamine and 35S-sulfate. [Show full abstract] carbazole sulfuric acid method and orcinol method and it was revealed that WF fibroblasts had a large amount of total uronic acid in the cell layer, the largest portion being the iduronic acid fraction. Electrophoretic analysis of the GAGS of WF showed an increase of production. Increased release into the medium and accumulation of pericellular glycosaminoglycans was observed in hyaluronic acid (HA), heparan sulfate + dermatan sulfate (HS + DS), and chondroitin sulfate (CHS) fractions. No specific accumulation of GAGS in the intracellular pools was observed.

Glycosaminoglycans were isolated from the four sections (tip, upper, middle and base) of the main beam of growing antlers of wapiti (Cervus elaphus) by papain digestion and DEAE-Sephacel chromatography. Chondroitin sulfate was the major glycosaminoglycan in each section of antler accounting for, on average, 88% of the total uronic acid. The yield of chondroitin sulfate liberated from the tissue ... [Show full abstract] was approximately 6-fold greater in the cartilaginous (tip and upper) sections than in the bony (middle and base) sections. This was consistent with the higher intensity of glycosaminoglycan staining with either Alcian Blue or Safranin-O. The majority (average 88%) of chondroitin sulfate was precipitated with 40 and 50% ethanol. The average molecular size of chondroitin sulfate determined by gel chromatography on Sephacryl S-300 tended to be greater in the 40% ethanol than in the 50% ethanol fraction. In either fraction, the molecular size of chondroitin sulfate was smaller in cartilaginous tissues than in osseous tissues of growing antler. In addition to chondroitin sulfate, the antler contained small amounts of hyaluronic acid, dermatan sulfate and keratan sulfate. The immunohistochemical study showed wide distribution of chondroitin sulfate, decorin, and keratan sulfate throughout the antler. On the other hand, keratan sulfate was more prominent in the cartilaginous sections than in the bony sections where the anti-keratan sulfate monoclonal antibody staining was seen in the osteoid tissue only.

[Show full abstract] into components by precipitation on a CPC saturated cellulose column, followed by fractional solubi-lisation of the CP-polysaccharide complexes by salt solutions of increasing concentrations.The fractions isolated were analysed by chemical and enzymatic procedures and were also refractionated by an analogous CPC micro procedure. Hyaluronic acid, heparan sulphate, chondroitin sulphates and dermatan sulphate were identified in all materials investigated. The galactosaminoglycans could not be successfully separated: the isometric chondroitin sulphates were quite inseparable, while a purified dermatan sulphate fraction could be obtained only at the expense of considerable loss of material. The results suggest a pronounced polydispersity of the aortic galactosaminoglycans with regard to chain length and/or degree of sulphation.It is stressed that glycosaminoglycan heterogeneity must be taken into account when attempts are made to identify these substances by means of the solubility characteristics of their CP complexes.

Polysaccharides of metaplastic mucosa and carcinoma of the gallbladder

[Show full abstract] In histochemical stainings it was found that in carcinomas and in the 2 specimens classified as group III (containing the most extensive metaplastic changes at disposal), the intracellular mucin was mainly neutral or nonsulfated acidic. The amounts of sulfated mucin were relatively insignificant. This mucin polysaccharide material was isolated and its composition was determined. It was observed to be large molecular (approximate molecular sizes 1 to 2 x 106), and to be composed of fucose, galactose, glucosamine, and galactosamine as well as small amounts of sialic acid. The basic structure of these polysaccharides is thus similar to that of normal sulfated mucin. The almost total absence of acid groups, however, causes the polysaccharide material in question to stain in a manner identical with neutral mucin when investigated with histochemical methods. The carcinomas also contained some sulfomucin; its proportion, however, was small as compared with the amounts of nonsulfated acid and neutral mucin in biochemical characterization. A small molecular polysaccharide fraction, assumed to originate in membrane bound glycoproteins, was isolated from the insoluble gallbladder tissue residue. The proportion of this fraction was larger in carcinomas than in normal material. This rise as well as the rise in the quantity of acid connective tissue polysaccharides is presumably due to the large number of cells in the carcinoma tissue as well as to fibrosis.

[Show full abstract] Distinct chemical differences were found between the total fractions of chondroitin sulfate from the three tissues. Within the total fractions a marked heterogeneity was also apparent.The turnover of the two glycosaminoglycans was studied by using radioactive sulfate as precursor. The guinea pigs were killed at eight intervals, ranging from 30 min to 40 days after injection of the isotope.Total fractions of chondroitin sulfate from rib, nasal septum and nucleus pulposus showed half-lives of about 80, 40 and 30 days, respectively. In addition, the existence of a second metabolic component with a faster turnover (half-life about 3 days) was indicated in chondroitin sulfate from all three tissues.Similarly, keratan sulfate from the rib contained a ‘slow’ component with a half-life of about 90 days and a ‘fast’ component with a half-life of 4 days. For keratan sulfate from nucleus pulposus only a single component was demonstrable; it had a half-life of about 30 days.Within the total fractions of both chondroitin sulfate and keratan sulfate from the three tissues studied, a considerable degree of metabolic heterogeneity was apparent between, and within, subfractions of differing molecular weight.

Fractionation and isolation of heparan sulfates using poly-L-lysine-Sepharose.

A simple procedure for the isolation of heparan sulfates from pig lung using a poly-L-lysine-Sepharose column is described.
