Possible roles of calcium and ammonium in the development of bitter pit in apple

ABSTRACT Apple trees (Malus pumila Mill. var. domestica Fuji/Malus prunifolia rootstock) showed a high susceptibility to bitter pit when supplyed with ammonium salt instead of nitrate (control) in the nutrient solution. When apple fruit was affected by bitter pit, a lower calcium as well as a higher nitrogen and ammonium-nitrogen contents was observed in the fruit flesh near the calyx end. The activity of the mitochondrial Ca2+-uptake of the fruit flesh near the calyx end was higher when the tree was grown with ammonium salt than when grown with nitrate. Both the activities of succinate: cytochrome c oxidoreductase and the mitochondrial Ca2+-uptake per g of tissue were higher in affected fruit than in healthy fruit. Each of chlorpromazine, N-(6-aminohexyl)-5-chloro-l-napthalenesulfonamide (W-7) and N-(6-aminohexyl)-l-naphthalenesulfonamide (W-5), calmodulin antagonists, was infiltrated into the fruit for 20 min under reduced pressure (about 1 × 104 Pa). Few days later, numerous bitter pit-like spots were observed in both fruit treated with W-7 and chlorpromazine, while only a few spots were observed after the infiltration with W-5, a less potent calmodulin antagonist. A possible mechanism for the occurrence of bitter pit is discussed.

    ABSTRACT: Crude Ca(2+)-activated protein kinase from membranes of apple (Malus domestica L. Borkh., Cox's Orange Pippin) fruit can be partially purified to yield a Ca(2+)-dependent protein kinase whose activity is apparently not regulated by calmodulin. The autophosphorylating catalytic subunit of this protein kinase shows a Ca(2+)-dependent mobility shift of approx. 10 kilodaltons (kDa) on sodium dodecyl sulphate-polyacrylamide gel electrophoresis; in the absence of added Ca(2+) or ethylene glycol-bis(β-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) its apparent molecular mass is approx. 50 kDa. The Ca(2+)-dependent protein kinase is inhibited by the calmodulin antagonists N-(6-aminohexyl)-5-chloro-1-naphthalenesulphonamide and trifluoperazine with IC50 values of approx. 45 μM and 15 μM, respectively. These similarities between the protein kinase and calmodulin indicate that the kinase may be a calmodulin-like protein.

    ABSTRACT: The study was conducted to determine induction of bitter pit in apples. Different forms and levels of nitrogen and levels of calcium were applied to water culture to examine bitter pit incidence and mineral contents of fruits and leaves. The NH_4-N group showed significantly higher nitrogen contents in both fruits and leaves than those of the NO_3-N group. However, calcium concentrations were decreased in the group of NH_2-N. The NH_2-N group showed the middle range in contents of nitrogen and calcium. Bitter pit incidence in the NH_4-N group was the highest among the treatments. Bitter pit incidence tended to be decreased as the nitrogen level reduced. As the calcium level increased, calcium contents in fruits and leaves were significantly increased, while those of nitrogen, N/Ca ratio, and bitter pit incidence decreased. Therefore, the results of this study suggested that bitter pit incidence was highly to nitrogen and calcium concentrations of fruits and leaves in apples.

    ABSTRACT: In order to establish the interactions of calcium sprays and exogenous propylene on the ripening of Anjou pears when either freshly harvested or chilled for 115 days, 2 levels of CaCl2 preharvest sprays and 2 levels of propylene (an ethylene analogue) were used postharvest. Freshly harvested fruits failed to ripen in air, while 50 μl/l propylene during ripening stimulated softening in all cases but failed to trigger any ethylene synthesis and resulted in a slow increase in respiration over a 30-day period. Treatment with 500 μl/ l propylene during ripening first accelerated softening, then initiated ethylene and CO2 production. Fully chilled fruit (115 days at −1 °C) when returned to ambient temperature ripened satisfactorily within 8 days regardless of the propylene treatment. Ethylene amounts produced after the chilling requirement was satisfied, were three-fold that produced at harvest. Calcium uptake was significant only when high concentrations were applied, but even this had only slight effects on ripening behavior. Although the controls and calcium treated fruits had the same firmness at harvest, the high Ca fruits showed consistently better firmness retention and slower increase in internal ethylene.
