Determination of Toosendanin Extracted from Traditional Chinese Medicine: the Fruit of Melia azedarach LC–FD Detection

ABSTRACT A rapid and selective reversed-phase liquid chromatography with fluorescence detection was developed and validated for the
determination of toosendanin in the fruit of Melia azedarach. After extraction, the extracts were analysed at 25°C using Eclipse XDB-C18 column (4.6mm×150mm; 5μm) and a mobile
phase consisting of methanol–water at a flow rate of 1.0mLmin−1. Detection wavelength was set at λEx=230nm, λEm=320nm. The linearity, accuracy, and precision of the method were good, and the method can be successfully used to investigate
the level of toosendanin component in herb samples with satisfactory results.

    ABSTRACT: Toosendanin (TSN) is a triterpenoid derivative found in Melia toosendan Sieb. Et Zucc (Meliaceae) or chinaberry. TSN present in the medicinal plants was first isolated and established by spectroscopic methods. In this report, high-performance liquid chromatography (HPLC) separation using columns of smaller particle size with tandem mass spectrometry (MS(n)) was used for the rapid determination of TSN in botanical extracts. A comparison of different fragmentation patterns shows that the results from positive and negative ion electrospray ionization (ESI)-MS(n) are complementary. The two modes can yield structurally significant information for the characterization and rapid identification of TSN in botanical extracts. The data obtained showed that MS(3) generated more characteristic ions that are useful for the identification of TSN in unknown samples. The separation of TSN was achieved with a water/acetonitrile gradient system using a short C18 reversed-phase column with small particle size (50 x 2.0 mm, 3.5 microm). With LC/MS, the quantitative analysis of TSN in the botanical extracts was done using external standard calibration and the method precision was found to vary from 4.3 to 7.6% (RSD, n = 5) on different days.

    ABSTRACT: Repellency, toxicity, and fecundity-reducing effects of bark extracts of Melia toosendan on three stored-product beetles, the rusty grain beetle, Cryptolestes ferrugineus (Stephens), the rice weevil, Sitophilus oryzae (L.), and the red flour beetle, Tribolium castaneum (Herbst), were investigated in the laboratory. Wheat kernels treated with extracts containing 75 and 3% toosendanin at concentrations from 0.05 to 0.4% (375-3000 ppm toosendanin), and from 0.5 to 2.5% (150–750 ppm toosendanin), respectively, repelled beetles by 50–98%, compared with controls. The test materials were more toxic to S. oryzae than C. ferrugineus and were not toxic to T. castaneum. The LC50 values for the extract containing 75% toosendanin for S. oryzae and C. ferrugineus after 6 weeks exposure were 675 and 1875 ppm toosendanin, respectively. Both extracts significantly reduced F1 adults of all three insect species in most treatments. Pure (98%) toosendanin caused only 4% mortality of C. ferrugineus and S. oryzae at a concentration of 0.15% (1470 ppm toosendanin) after 2 weeks, but significantly reduced the F1 adults compared with controls. This study indicates that a natural grain protectant based on M. toosendan extracts may be feasible.

    ABSTRACT: The effects of a specific presynaptic cholinergic antagonist, toosendanin, on memory formation following a passive avoidance training experience in day-old chicks was investigated. Bilateral injection of toosendanin into the neostriatal/hyperstriatal region of the chick forebrain produced memory impairment in a dose-dependent manner. Retention deficits were apparent from 20 min following training in chicks treated with toosendanin, regardless of the injection time relative to training. Chicks that received injections of the drug at corresponding times prior to retention tests showed normal retention levels, suggesting that toosendanin has no effect on performance and memory retrieval. These results indicate an involvement of cholinergic transmission during an early stage of memory formation.
