• Repairing of oxidative damage to mesenchymal stem cell in rats and anti-lipidperoxidation by Plastrum Testudinis ethanolic extract
ABSTRACT Objective】To study Plastrum Testudinis ethanol extract’s activities of repairing oxidative damage to mesenchymal stem cell(MSC) and its anti-lipidperoxidation.【Methods】(1) MSCs were isolated from rats by density gradient and cultured , then identified by CD44. The model of MSC’s oxidative damage was created by using hydrogen peroxide(H2O2) for 3 hours. The activity of repairing oxidative damage to MSC was determined by MTT method, and the extract’s concentration varied ,including 0.166,0.833,1.66,3.32,4.98mg/mL. (2) 50 SD rats were random divided 5 groups in average. The extract group: three doses(0.01、0.03、0.09g/(kg•d) were given by i.g.; The positive control group: vit.E 200mg/(kg•d) was given by i.g.; The control group: distilled water 200mg/(kg•d) was given by i.g..The drug was given once a day and lasted for 7 days .Then, MDA and SOD in rats’ liver were measured by TBA method and nitrous salt method.【Results】(1) There were distinct difference between H2O2 group (1:10,V/V) and Plastrum Testudinis ethanol extract group in promoting optic density of MSC（P<0.01,P<0.05）; (2) Compared with the control group, the level of MDA of extract group was greatly lower （P<0.01）, the activity of SOD was greatly higher（P<0.01,P<0.05.【Conclusion】Plastrum Testudinis extract can repair oxidative damage to mesenchymal stem cell(MSC) from rats, and reduce lipid peroxidation.
