Central nervous system expression of HIV-1 Gp120 activates the hypothalamic-pituitary-adrenal axis: Evidence for involvement of NMDA receptors and nitric oxide synthase

ABSTRACT The impact of HIV-1 expression in the brain on the development of AIDS is unknown. In the present study, we examined the hypothalamic-pituitary-adrenal (HPA) axis in a transgenic model in which expression of the HIV-1 envelope glycoprotein gp120 induced central nervous system (CNS) damage similar to that seen in HIV-1-infected patients. Compared with nontransgenic littermates, gp120 transgenic mice showed significant increases in plasma corticosterone and adrenocorticotrophic hormone (ACTH) levels and pituitary ACTH content. To determine whether this activation of the HPA axis could be mediated by ACTH secretagogues, the effect of recombinant gp120 on the release of these factors from hypothalamic slices was investigated in vitro. Recombinant gp120 induced release of the ACTH secretagogue arginine vasopressin from nontransgenic hypothalamic slices in a calcium-dependent fashion. This effect was inhibited by antagonists of N-methyl-D-aspartate (NMDA) receptors or of nitric oxide synthase (NOS), suggesting a role for NMDA receptor stimulation and NOS activity. Further evidence for a role of free radicals was obtained from bigenic mice coexpressing gp120 and the free radical scavenger human copper/zinc superoxide dismutase which showed normal corticosterone levels. This might relate to superoxide dismutase-mediated scavenging of superoxides generated by NOS. These findings demonstrate that CNS expression of a viral envelope protein can activate the HPA axis and thereby alter peripheral levels of immunomodulatory hormones.

    ABSTRACT: Immune dysfunction resulting from HIV infection is a major health threat. As HIV-infected persons survive previously life-threatening infection through the use of effective medical therapies, undernutrition, its immune dysfunction and heart disease have become critical issues in the health care of longer-term survivors. Changes in oxidation and the resulting immune modulation affect their host defenses. The potential role of oxidation and immune dysfunction in HIV-induced heart disease is reviewed. Antioxidant is becoming accepted as interventions in the early and ongoing treatment of HIV disease. Nutritional and hormonal therapy, in coordination with other medical interventions, should extend and improve the quality and quantity of life for individuals infected with HIV. Research about the relationship between nutrition/hormone and development of AIDS is essential for understanding its mechanisms of immune dysfunction and heart disease, and for determining the effectiveness of nutritional and hormonal therapy. The mechanisms and consequences of oxidation and immune dysfunction in HIV/AIDS patients are reviewed with special emphasis on heart disease.

    ABSTRACT: Carnivore extinctions frequently have cascading impacts through an ecosystem, so effective management of ecological communities requires an understanding of carnivore vulnerability. This has been hindered by the elusive nature of many carnivores, as well as a disproportionate focus on large-bodied species and particular geographic regions. We use multiple survey methods and a hierarchical multi-species occupancy model accounting for imperfect detection to assess extinction risk across the entire carnivore community in Ghana’s Mole National Park, a poorly studied West African savanna ecosystem. Only 9 of 16 historically occurring carnivore species were detected in a camera-trap survey covering 253 stations deployed for 5469 trap days between October 2006 and January 2009, and our occupancy model indicated low overall likelihoods of false absence despite low per-survey probabilities of detection. Concurrent sign, call-in, and village surveys, as well as long-term law enforcement patrol records, provided more equivocal evidence of carnivore occurrence but supported the conclusion that many carnivores have declined and are likely functionally or fully extirpated from the park, including the top predator, lion (Panthera leo). Contrary to expectation, variation in carnivore persistence was not explained by ecological or life-history traits such as body size, home range size or fecundity, thus raising questions about the predictability of carnivore community disassembly. Our results imply an urgent need for new initiatives to better protect and restore West Africa’s embattled carnivore populations, and they highlight a broader need for more empirical study of the response of entire carnivore communities to anthropogenic impact.

    ABSTRACT: The hypothalamic pituitary adrenal (HPA) axis is the most common of the endocrine lines/axis' to be affected by HIV infection. There are multiple factors that contribute to this HPA axis dysregulation. Direct invasion of the various organs in the axis can be either by opportunistic infections or infiltrative diseases. The soluble factors or cytokines released during viral infection and the chronic inflammatory state that follows, also contribute to these alterations. The actions of these cytokines released by the immune response can both activate the HPA axis and cause a glucocorticoid resistant state. Further, many of the anti-retroviral and other medications used to treat HIV infection can contribute to HPA axis dysfunction. While the diagnosis and treatment of endocrine dysfunction is the same as in any other patient, management pathways may be quite different. While some may be adaptive responses, life threatening adrenal insufficiency can also be present. It is important the latter be picked up expeditiously and treated promptly to avoid mortality.
