Polyamine distribution profiles in the eighteen genera phylogenetically located within the Flavobacterium-Flexibacter-Cytophaga complex.
ABSTRACT Cellular polyamines of eighteen genera belonging to the Flavobacterium-Flexibacter-Cytophaga complex were analysed by ion exchange liquid chromatography. Homospermidine was the major polyamine in the genera Bergeyella, Riemerella, Ornithobacterium, Weeksella, Capnocytophaga, Polaribacter and Psychroflexus belonging to the family Flavobacteriaceae. In the family Spirosomaceae, Runella, Spirosoma and Flectobacillus species contained spermidine whereas Cyclobacterium species contained homospermidine. Within a divergent cluster, Haliscomenobacter and Lewinella species contained spermidine whereas Saprospira grandis contained agmatine alone. The major polyamine of Chitinophaga and Sporocytophaga species was homospermidine. Flexithrix dorotheae contained spermidine. Microscilla marina, the type species of the genus Microscilla, contained spermidine and cadaverine. However, 'Microscilla sericea' contained homospermidine, 'Microscilla furvescens' contained spermidine, and 'Microscilla arenaria' lacked all polyamines. Polyamine profiles serve as a phenotypic chemotaxonomic marker for the reclassification of the genera belonging to the complex.

ABSTRACT: A Gram-reaction-negative, strictly aerobic, non-pigmented, non-gliding, rod-shaped bacterium, designated strain BY4T, was isolated from freshwater. Cells were catalase- and oxidase-positive and indole was produced. Phylogenetic analyses based on 16S rRNA gene indicated that strain BY4T belonged to the family Flavobacteriaceae and showed 91.6-95.9 % sequence similarities to the most closely relatives. The major respiratory quinone was MK-6 and the major polar lipid was phosphatidylethanolamine. The major polyamine was homospermidine and the major fatty acids were iso-C15:0, iso-C17:0 3-OH and summed feature 3 (C16:1ω7c and/or C16:1ω6c). The DNA G+C content was 30.0 mol %. Based on the phenotypic, phylogenetic and genotypic features, strain BY4T is suggested to represent a novel genus and species within the family Flavobacteriaceae, for which the name Chishuiella changwenlii gen. nov., sp. nov. is proposed. The type strain is BY4T (= CGMCC 1.12707T = JCM 19633T). Based on the data collected from previous and present studies, it is proposed to reclassify Wautersiella falsenii to the genus Empedobacter as the new combination Empedobacter falsenii comb. nov. ABSTRACT: A Gram-reaction-negative bacterium, strain IMMIB L-12475(T), was isolated from blood cultures of a human with septicaemia. The yellowish orange pigmented strain contained flexirubin pigment. Phylogenetic analysis based on 16S rRNA gene sequence revealed that strain IMMIB L-12475(T) belonged to the family Flavobacteriaceae, forming a distinct phyletic line that is distantly related (79.1-89.4% sequence similarity) to described genera of this family. Membership to the family was confirmed by a fatty acid profile consisting of branched-chain and 3-hydroxy fatty acids with major amounts of iso-C(17:0) 3-OH and iso-C(15:0), by the presence of menaquinone MK-6 as the only respiratory quinone and a polyamine pattern that contained sym-homospermidine as major component. The phospholipids consisted of phosphatidylethanolamine and an unknown phospholipid. The genomic DNA mol% G+C content was 45.6%. The distant phylogenetic position as compared to other representative of the family and the significant phenotypic properties such as pigment composition, morphology, and physiology support the proposal of a novel genus and species Cruoricaptor ignavus gen. nov., sp. nov.
