雞傳染性喉頭氣管炎的診斷,除靠疫學、臨床症狀、解剖及組織病理學病 變外,尚須配合一種以上的實驗室檢驗才能加以確診。本實驗開發了以聚 合酵素連鎖反應來偵測傳染性喉頭氣管炎病毒核酸的技術,以期能快速診 斷此病。雞傳染性喉頭氣管炎野外病毒株(埔里株)的0.63 kb HindⅢ片斷 經定序後,設定 1對外側引子進行聚合酵素連鎖反應可得一長度為516 bp 之特異性片斷,其敏感度達1 pg之病毒核酸;取部分產物以另 1對內側巢 式引子進行巢式聚合酵素連鎖反應可得另一長度為260 bp之特異性片斷, 其敏感度則高達1 fg (約相當於 6套病毒基因體所含核酸)。以 6株不同 的野外株及疫苗株病毒核酸為模板進行反應,可證明其能廣泛適用於各病 毒株;再以雞胚胎腎臟細胞、馬立克病毒血清型1、2型、火雞皰疹病...
