[more]1) Propogate the Lactobacilli MRS media for 3 days using a shaker incubator. 2) The culture media is subjected to centrifugation at 4°C for 20 min. 3) Preciptate the Cell Free Supernatent(CFS) using 85% ammonium sulphate. 4) Again centrifuge the cell free supernatent at 4°C for 35 min. 5) Go for the appropriate purification technique, usually ion- exchange chromatography is preferred. In case of non-lanthionine bacteriocins, ion exchange chromatogrpahy using a octyl-sepharose CL-4B (Octyl Sepharose™ CL-4B is a separation medium for hydrophobic interaction chromatography (HIC). Fenugreek seeds were enriched by soaking them in Cr(3) picolinate solution for 24hr under controlled temperature and humidity.Then the Cr(3) fenugreek seeds was analysed quantitatively for alkaloids, saponins and Cr(3).The quantification of alkaloids and saponins was done using solvents.Methanol:water(70:30 by volume) gave the max yield of alakloids.The quantity of alkaloids and saponins present in the Cr(3) enriched fenugreek seeds remained the same as in the control(normal fenugreek seeds).
