[more]Hi Bram, for transfecting human monocytes and generally suspension cells, you can try a viral approach in combination with ViroMag reagent (Magnetofection). ViroMag would increase viral infectivity and transduction efficiency in any suspension cells such as PBL and PBMC and ViroMag plus a virus have been successfully used on primary T cell and several citations reports have been published (Sacha JB., Nat Protoc. 2010 ; Sakhdari A., PLoS One. 2012) If you prefer a non viral approach you can try with a lipid based reagent. DreamFect Gold reagent is performant on immortalized Lymphocyte cells such as THP-1 (Pakula R., Glycobiology. 2007; Vanpouille C., J Biol Chem. 2007; Wenzel M., J Immunol. 2012).
