The concentration of P... [more]I have tried to make the PEI-DNA complex with mole ratio of PEI and DNA as 1:1, 2:1, 4:1, 8:1, 12:1, 16:1, 20:1, 24:1 and 28:1. The concentration of PEI and DNA is 2mg/ml and 2mg/ml, respectively. I found when I mingle them, the solution is still clear. However, after a while(not more than 15 min), most of them would be turbid in different degrees. When I am evaluating the diameter of the particles, they are usually more than 300nm and it may be increased with time if you try to evaluate more times. I have checked everything I used and I could promise that nothing is contaminated. But how could the solution get turbid?By Haoyang Jiang · Nanjing UniversityFollowing

However, when I checked them under the confocal in 1 and 2 days after the transfection, I could see a lot of cells with GFP overexposure although some other cells have more nomal and dim green light.
