[more] The discovery of micro RNA (miRNA) in 1993 by Ambros and colleagues has a huge influence in pathogenesis theory, diagnosis and treatment approach of some diseases. Some studies have conducted to seek the association alterations of miRNA expression to incidences and severity of preeclampsia (PE). We have reviewed some studies that conducted to seek the association of miRNA and PE and we discussed the role of various miRNAs in PE pathogenesis. In summary, we have shown that many researchers have given evident that the different placental and plasma miRNA expression is associated with PE. Some studies also identified the novel candidate of miRNAs (and their pathways) that may be of etiologic relevance in the pathogenesis of PE. Base on review, specific miRNA have a role to down regulate of anti apoptosis genes, regulate angiogenics growth factors such as angiogenin, vascular endothelial growth factor (VEGF) B (VEGF-β), cysteine-rich 61 (CYR61), Placental growth factor (PlGF) and VEGF-A that have a role in angiogenesis. miRNA also have a role in survival, migration, and capillary tube formation of HUVEC by targeted of c-kit. Some miRNAs target genes that participate in immunologic dysfunction, cell adhesion, cell cycle, and signaling. miRNA also have a roles in endothelial cell response to hypoxia, cell differentiation, and survival. A miRNA influence calcium signaling through negative regulations of the calmodulin-coding mRNAs, Mef2a and Gata4, mainly in smooth muscle cells that contribute to PE pathogenesis. These investigations provide novel targets for further investigation of the pathogenesis of PE and these differential miRNAs may be potential markers for the diagnosis and provide a potential therapeutic target for PE. Further investigations on posttranscriptional regulation in PE to evaluate biologic effects of identified miRNAs (including confirmations of miRNA and target gene interactions) are needed.

[more] The honey is known by people as a medicine for threatment several deseases, but until now the used of honey has not naximun jet . The purpose of this research is to determine the effect of honey on microscopic appearance of the white pulp part of spleen Mus musculus after infected by Pseudomonas aeroginosa. Research design used was posttest only control group design. Three groups of M. musculus strains DJ, male, 1.5 months were used four times each. Doses of honey given were 0.8 ml, 0.4 ml, and 0.2 ml per day for each group. Honey diet was conducted over 20 days. P. aeruginosa injected intraper­itoneally into each animal with a single dose of 0.1 ml of 2 Mc Farland. M. musculus were incised and spleen histology preparations with HE dye were made, then diameters of the white pulp spleens were measured using a microscope by 100x magnification. From the research, there was no significa­nt difference in the diameter of the white pulp spleen between group of 0.8 ml dose of honey and group of 0.4 ml dose of honey (p = 0.164.) There was significant difference of white pulp spleen diamet­er between group of 0.8 ml dose of honey and group of 0.4 ml dose of honey (p = 0.001). There are also significant difference of white pulp lien diameter between group of 0.4 ml dose of honey and group of 0.2 ml dose of honey (p = 0.000). These results showed that honey can improve the body’s immun­e system by increasing the activity of white pulp spleen of M. musculus. However, the mechanism can not be determined. Therefore, it’s necessary to determine the cellular and molecular mechanism­s of how honey increases the body’s immune system.
  • Combination of Immunoglobulin G3 and Eosin B: Plasmodium falciparum merozoite Invasion Inhibitor

Therefore, ... [more] Malaria caused by Plasmodium result 300-900 million morbidity and 1-3 million mortality annually. The use of anti-malarial drugs such as kloroquin, atovaquon, mefloquin, proguanil, and pirimethamin has encountered a problem because the emergence of P. falciparum resistant to these drugs. Therefore, it is necessary to search for anti-malarial drugs that are not easy to resistance. The study ever done shows a good opportunity to use immunoglobulin G3 (IgG3) and eosin B as an anti malarial which has mechanism to inhibit invasion process of P. falciparum into erythrocyte. IgG3 inhibits merozoites invasion by increasing resistance to erythrocyte binding antigen 175 (EBA-175) in regions III-V referred to EBA peptide 4, region II, and regions C and F segments dimorphic. This will inhibit the binding of EBA-175 to erythrocytes so that it will inhibit merozoites invasion into erythrocyte. IgG3 also inhibits several merozoite-associated surface proteins (MSP) that are involved in the process of merozoites invasion. While, eosin B has the mechanism to inhibits several enzymes, including dihydrofolate reductase-tymidylate syntase (DHFR-TS), Plasmodium falciparum glutathione reductase (PfGR), Plasmodium falciparum thioredoxin reductase (PfTrxR), human glutathione reductase (hGR), and human thioredoxin reductase (hTrxR) on ring-stage that will cause phagocytosis. Based on the mechanism of IgG3 and eosin B, thus both of these components have an opportunity as an anti-malarial drug combination in the future

    The aim of this study was to identify the level of health care worker`s (HCW) stigmatized and discriminatory (S&D) attitude towards people living with HIV (PLHIV) and the factors that influenced this attitude. This research was conducted at the Dr. Zainoel Abidin General Hospital (RSUDZA) in Ban... [more] The aim of this study was to identify the level of health care worker`s (HCW) stigmatized and discriminatory (S&D) attitude towards people living with HIV (PLHIV) and the factors that influenced this attitude. This research was conducted at the Dr. Zainoel Abidin General Hospital (RSUDZA) in Banda Aceh province of Indonesia. A cross-sectional study design was adopted for this research. Eighty nine HCWs were included in this study and they were selected purposively. Pearson correlation analysis, analysis of variance or independent sample t test analysis was used according to the type of data. We found that the level of S&D attitude towards PLHIV in RSUDZA is high. Based on the Least Significant Difference Test, the average score of the answers between nurses and general practitioners and between nurses and medical students was significantly different(p=0.05). Pearson correlation analysis showed that the levels of S&D attitude towards PLHIV was positively correlated with age (r=0.219, p=0.04) and irrational fear of HIV transmission (r=0.352, p=0.001) and negatively correlated with knowledge about HIV/AIDS (r=-0.336, p=0.001). Pearson correlation analysis also found that knowledge of HIV/AIDS negatively correlated with the irrational fear of HIV transmission (r=0.382, p=0.000). In addition, the level of S&D attitude towards PLHIV was also associated with marital status of HCWs (p=0.020). Gender, education level, religion and the importance of religion in HCW lives wasn`t significantly affect to the level of S&D attitude towards PLHIV in HCW. We concluded that the factors that influence the level of S&D attitude towards PLHIV among the HCW are age, marital status, knowledge of HIV/AIDS, irrational fear of HIV transmission and HCWs occupations. To reduce S&D attitude towards PLHIV among the HCWs, we recommend introducing some program to increase knowledge of HIV/AIDS.

The evidence that endogenous mediators actually mediate the individual’s response to infection has led to various approaches to assess the individual’s contribution to the course of the disease.... [more] Sepsis and its sequelae are still a major cause of morbidity and mortality in intensive care units (ICU). The evidence that endogenous mediators actually mediate the individual’s response to infection has led to various approaches to assess the individual’s contribution to the course of the disease. The role of an individual’s genetic background and predisposition for the extent of inflammatory responses is determined by variability of genes encoding endogenous mediators that constitute the pathways of inflammation. Pro- and anti-inflammatory responses contribute to the susceptibility and outcome of patients with systemic inflammation and sepsis. Therefore, all genes encoding proteins involved in the transduction of inflammatory processes are candidate genes to determine the human genetic background that is responsible for interindividual differences in systemic inflammatory responses. Polymorphism of TNA α, TNFB, IL-10, IL-6, IL-1β, IL-1RN, HMGB1, TLR, PAI-1, DEFB1, HSP and MMP-9 has contribution to susceptibility and outcome of sepsis in some research. Examination of the association between genetic polymorphisms and sepsis promises to provide clinicians with new tools to evaluate prognosis, to intervene early and aggressively in treating high risk persons, and to avoid the use of therapies with adverse effects in treating low risk persons. Genomic information may be useful to use to identify groups of patients with a high risk of developing severe sepsis and multiple organ dysfunctions.

[more] This research conducted to know the impact of masturbation and porn movie to anxiety neurosis and memory recall of teenager. This research was conducted to 217 students at eleven Senior High Schools in Banda Aceh, Aceh Besar and Pidie Region in Nanggroe Aceh Darussalam. This research uses the incidental sampling method. To collect all of the data used closed ended questioner instrument and answer by respondent directly. In this research used statistical proportional percentage with description analysis. From the research we found that 41.96% of respondent have watched the porn movie and 45.62% of respondent have masturbation two times in a week. About 67.28% respondent got anxiety neurosis if they have masturbation and 53.92% that respondent had decrease of recall ability. Masturbation is an activity that is come from porn movie, this is dedicated to discard unstoppable libido. The unstoppable libido can cause the crisis situation in the teenager that induces of internal confliction between unstoppable libido and religion or culture. If the libido become huge and they have masturbation, the guiltiness appear as the respect of their conscience to what they has done. This Internal conflicti­on will disturb the learning process. Endogenic factor of learning process which is disturbed by the influence of porn movie and masturbation are the concentration, motivation and emotion. Further more, all of these will disturb coding process, information storage process and memory recall of the studen­t.

Nucleoside Reverse Transcriptase Inhibitor (NRTI), Non-Nucleoside Reverse Transcriptase Inhibitor (NNRTI) and Protease Inhibitor that used in last therapy have serious problem because they can increase HIV-1 mutation frequency and resulting in drug resistant HIV-1 Variant. Because these problems, effort to find a new antiretroviral drug is a solution to solve this problem. Beside the last antiretroviral drug target site, one of HIV-1 life cycle phase that possible to block with new antiretroviral drug is fusion phase between HIV-1 particle and human CD4 cell (HIV-1 entry inhibitor). T-20 is one of HIV entry inhibitor that have used worldly, but single regiment treatment have big chance produce HIV resistant, thus combination drug treatment needed, beside that combination drug that have synergetic action mechanism useful to get more effective therapy. One of HIV- 1 entry inhibitor drug development that have big opportunity is Tetravalent CD4-Immunoglobulin G2 (PRO 542), protein fusion between variable subunit of light chain and heavy chain IgG2 and D1D2 domain of human CD4 cell. PRO 542 inhibit attachment of viral gp120 in CD4 receptor, so viral gp120 can't interacted with CXCR4 or CCR5 co-receptor, finally inhibit Phe-43 cavity formation, that very important in HIV-1 genetic material transfer to CD4 cell target. Because target site of PRO 542 is structural component of HIV-1, gp120, so the resistant pathway for PRO 542 is not easy occurred. Combination T-20 and PRO 542 can reduce therapy dose up to 10 times, beside that this combination can prevent drug resistant because both of them have synergetic action mechanism - inhibit HIV-1 entry to CD4 cell. Because of this synergetic action mechanism, this combination can give effective and efficient therapy in HIV/AIDS patient. But furthermore research about drug resistant to this combination is needed. Beside that clinical trial to determine dose therapy for this combination regiment and PRO 542 site effect to human cell are needed.
