Risk Factors In Adolescent On Arteriosclerosis: An Analysis Using Data Mining And Docking Techniques

[more] Aims: Data related to arteriosclerosis has been collected from different hospitals from Visakhapatnam, India. Data has to been analyzed for future experimentations so that mechanism of metabolites on aging diseases such as arteriosclerosis can be cured. Methods:Data related to arteriosclerosis has been collected from different hospitals in Visakhapatnam during January and February 2011. Analysis of this data has been done using machine learning approaches. Docking studies related to prescribed drugs has been conducted using docking software. Results: The results showed that the arteriosclerosis occurs mostly to the old people (60-69 years). The data analyzed has been done through data mining techniques which showed 92.9% accuracy using BayesNet. From the data collected, the drugs which are being used are docked with a diseased protein which causes atherosclerosis, showed Betaloc can be an anti-arteriosclerotic effective drug. Arteriosclerosis is a metabolic syndrome and genitical disease, occurring more in the persons who prefer tea rather than coffee. Eighteen percent of the persons are predicted with family background. Conclusion: The present results showed that Age, height, gender, weight, family history, tea, coffee, number of times tea/coffee consumption, blood test, cholesterol, diabetes, obesity, drinking and smoking are the factors responsible for arteriosclerosis based on data analysis and machine learning approaches. Clinical research has to be conducted further in this direction.

[more] Comparative genome analysis are playing important role in genomics, where the scientific studies can provide the relationship of two or multiple organisms. Both HIV-1 and HIV-2 predicted nine genes in entire genome. HIV is a nano particle (sometimes living organism) has molecular information which can use host machinery in the formation of proteins such as gag polyprotein, gag-pol polyprotein, vif, vpr, tat, vpu, gp160, envelope glycoprotein and Nef located in various regions of the genome. Vpu is not predicted in HIV-2. The three dimentional molecules can further be used for the identification of drug targets for the control of diseased molecules located in HIV.

In this point of view, eight viral sequences of H1N1 were downloaded from NCBI Tax Browser and scanned against complete genome of H1N1 for the presence of possible viral inserts in SARS genome. The results from the computational analysis provided that H1N1 virus resulted in viral segments inserted in the intron and exon regions of SARS genome. [more] In the present decades, science and scientific methods are exploring great inventions and much of the biological information, may be answered by the end of 21st century. The present research has been conducted to identify fungal species on spider and applied image analysis technique. During rainy and winter seasons, due to dramatic decrease in temperatures, fungi grow on the spiders as white colonies. Staining of these white, mushroom like mycelium has been done and identified two organisms, Nomuraea rileyi and Nomuraea atypicola. The fungal spores are infecting spiders and also other insects, through intake of food or entering into the body by respiratory track. An image analysis technique provides much clarity in microscopic images of fungi on degrading spiders taken by Olympus camera (2048X1536 pixel).

The present studies provided genomic and proteomic studies of SARS virus in application of computational tools. Genomic sequence of SARS virus (29,751 bp) from NCBI database predicted eleven potential genes. The sequences shown relationship with Porcine hemagglutinating encephalomyelitis virus and RNA murine hepatitis virus. The results also predicts that acyltransferase family proteins of Lactobacillus acidophilus and nucleocapsid of SARS coronavirus are extremely basic in nature and spike glycoprotein of SARS coronavirus predicted acidic in nature.

Enough evidence to support the claim of this problem can be provided by data mining and machine learning approaches. The data has been explored from UCI Machine learning repository and weka version 3.2 has been used as tool in the present research. Breast cancer, Embryonal Tumour on central nervous system, Leukamia, lung cancer and Lymphoma datasets provided good results with random forest method. Various attributes such as cell size, bare nucleoli, cell shape, clump thickness, marginal adhersion, genes 1637x, 1731x, 1062x, 3950x, and 3328x has shown association with cancer.

Granulocyte macrophage colony stimulating factor has shown good activity on Gag polyprotein (R.value 20.8). IGG2A intact antibody has shown good activity against pol polyprotein. Interferon alfa2B has shown better activity, forming complex with pol polyprotein (R Value 5.6, E,Total -573.81). IGG2A antibody has also shown better activity in deactivating envelop protein of ALV (R=8.8, E.Total = -496.11). Vindesine, altretinoin, Imatanib, Etoposide, Docetaxel, Flavopiridol has shown good activity against Gag, Poly, Env proteins of ALV.

HHEX is a gene significantly involved in developing D2M. .The metabolic pathway provides the information regarding the molecules responsibe for the cause of D2M. The molecular docking predicts the lowest binding energy using Emin values. All the drugs has shown negative energies, predicted that all the ligands act on hhex. Based on R-value and energy value in Hex,, Glimepride, Vitamin E and Acarbose showed better results. Based on GEMDOCK energy values, Glibenclamide and Mazindol predicted better results.

In Vitro Regeneration of the Medicinal Herb, Evolvulus Nummularius L. From Shoot Tip and Flower Explants

[more] Indirect shoot regeneration of E. nummularius was achieved by culturing terminal buds and flower on MS medium supplemented with various auxins (IAA and 2, 4-D) and Cytokinins (BA and Kn). Present protocol includes devising effective sterilization methods in treatment of explants. Weekly sub culturing to the media with same composition produced better results. Enormous amount of calli having regenerative potential were formed from the explants when cultured on MS medium supplemented with 1 mg/l 2, 4-D+1 mg/l BA. Different concentrations and combinations tested on MS medium containing 3 mg/l BA+ 0.5 mg/l IAA produced highest number of shoots per gram calli. Rooting was more at concentration of 1 mg/l IAA. The plantlets have shown eighty percent survival rate while transferring from in vitro to natural environment.

[more] Antithymocyte globulins (ATG) are effective therapies in aplastic anemia but their mechanism of action is undefined. The present research on ATGs globulins extracted from various precipitation reagents using SDS PAGE showed that the standards and markers at 52 KD are similar. The standards at 31 KD is similar. Extracts of poly ethylene glycol and Ammonium sulphate are similar between 52 KD and 98 KD. A molecular weight marker has got similarity with ATG standard. ELISA results have provided positive result, showing yellow coloured appearance. Hence a positive result with ATG polyclonal antibodies against T cell suspension antigens has been presented using SDS PAGE and ELISA.

Computational techniques have revolutionerized molecular modeling and most of the calculations cannot be performed without the use of a computer. Genome of CHIKV on submission to BLASTP is predicted with Helicase, peptidase_C9, Macro pep, RDRP_2 superfamilies in protein 1. Peptidase S3, Alpha E1, E2 and E3 superfamilies are predicted in Protein 3. Protein 2 is predicted relationship with glycoside hydrolase family 48 [Catenulispora acidiphila], hypothetical protein PEPMIC_01586 [Parvimonas micra], PAS/PAC sensor signal transduction histidine kinase [Frankia symbiont of Datisca glomerata], protein HOTHEAD [Zea mays], tuftelin [Macaca mulatta] and hypothetical protein [Trifolium pratense]. DENV-1 and DENV-3 are related with deviation in DENV-2 and DENV-4. CHIKV is closely related to DENV-4 in serotypes of DENV. CHIKV and DENV are closely related viruses with 95% similarity and 45% conserved sites in genomes. Hygienic conditions are required to stop spreading of viral diseases like CHIKV and DENV. The viral spread in Visakhapatnam regions provided a clue to spread of CHIKV in absence of pigs as reservoirs. The spread of disease is mainly due to the vector mosquitoes, principally Aedes aegypti and Aedes albopictus.

[more] The anti-apoptotic proteins play a vital role in regulating the apoptotic signaling. The inhibition of apoptosis process can be mediated through these anti-apoptotic proteins. Understanding the signaling of apoptosis in cancer state of a cell leads to a new therapeutic approach by inhibiting the anti-apoptotic proteins. Once the anti-apoptotic proteins are deactivated, it leads to the proper activation of apoptosis in cancer cells which leads to the death of a cancer cell. Hence, in this work the anti-apoptotic proteins were identified through literature and their tertiary structures were predicted using automated servers named CPH Models 3.0 & 3D Jigsaw. Some of the structures are directly retrieved from PDB. The respective ligand molecules were retrieved from Drugbank. The docking studies were performed between selective anti-apoptotic proteins & their respective ligands using Molegro Virtual Docker (MVD) form which higher docking rerank score is taken in to consideration as an effective inhibitor / ligand molecule to the anti-apoptotic protein.

In the present work, data is obtained from the Oxford Dataset and classification techniques such as CART, Random Forest, LMT, and Naïve Bayesian were used. The result predicted that Random forest method using training dataset outperforms the remaining methods. The random forest method using training dataset have less value of absolute relative error. Relative absolute error of LMT is high for cancer survival dataset. Value of absolute relative error is greater than 50% for almost all the algorithms except for random forest method using training dataset.

Different species of animals age at radically different paces and occurrences of diseases. Drug designing approaches related to an aging factor, Type 2 Diabetes mellitus has been reported in the present study. Resveratrol and human Sirtuins (1 to 6) are docked against Type 2 Diabetes mellitus HNF-1a motif with PDB ID: 2GYP using Hex 5.1. Resveratrol and Sirtuin 6 shown good results and can use as anti-aging drugs against Type 2 Diabetes mellitus.

[more] Study of antibacterial activity of hexane, chloroform and methanol extracts whole plant extract from Mucuna pruriens leaves has been evaluated, invitro against Erwinia carotovora, Pseudomonas syringae, Pseudomonas marginalis, Pseudomonas aeruginosa, Xanthomonas campestris. Study of antifungal activity of whole plant extract of Mucuna pruriens against Curvularia lunata, Fusariumoxy sporum, Pencillium expansum, Rhizoctonia solani, Tiarosporella phaseolina, Ustila gomaydis. All extracts exhibited antimicrobial activity against different species of bacteria and fungi. The methanol extract of Mucuna pruriens showed highest antimicrobial activity against all the bacterial and fungal species tested. Whereas chloroform extracts showed moderate antimicrobial activity against both bacterial and fungal species. The results of present study showed that Mucuna pruriens plant may be a good candidate as antimicrobial agent.

[more] Bioinformatics and computational biology involve the use of techniques with the goal of analyzing the sequence of Discopyge ommata to identify the genes. A marine ray Discopyge ommata gene (5'-nucleotidase) is used in the first cycle to search for predicting the locations and exon-intron structures of genes in genomic sequences from Genscan. The protein predicted more number of Glycine with divergence value is 328.67. More number of adenine is predicted in the gene of Discopyge ommata. The predicted protein of marine ray Discopyge ommata shows more number of aliphatic compounds and is closely related to Xenopus laevis (African clawed frog), Danio rerio (zebrafish) and mouse with Calcineurin-like phosphoesterase.

[more] Edible mushrooms are the fungi fruiting bodies which have high protein content along with several vitamins and minerals. Mushrooms have medicinal value in inhibiting tumour growth and enhancing immune system. The antimicrobial activity of 90% ethyl acetate and antioxidant activity of fruiting bodies of mushrooms of four edible mushrooms were investigated. The antimicrobial activity of four mushroom samples was tested against different bacteria using agar well diffusion method with ampicillin as positive standard reference to determine the sensivity of the bacterial strains. Two mushrooms samples showed significant inhibitory activity against Staphylococcus aureus which is a normal inhabitant in humans. The antioxidant activity, peroxidase, number of ascorbate oxidase units and catalase activity were significant more with Agaricus bisporus than the other edible mushrooms used in the study. All the activities measured are to find out for the presence of reactive oxygen species which play an important role in cell death and signal transduction.

[more] Huntington's disease results from genetically programmed degeneration of nerve cells, called neurons, in certain areas of the brain. This degeneration causes uncontrolled movements, loss of intellectual faculties, and emotional disturbance. The disease is caused due to the mutant huntingtin protein for which the function is unclear. This protein interrupts the other interacting proteins activity resulting in abnormal functioning of the nerve cells. To deduce the exact function of huntingtin protein, an attempt is made to determine the structure of Htt protein which is very large in length, through automated modelling server and phylogeny analysis as well structure alignment studies were performed using CLC Workbench & MVD software respectively for inferring the conservativity of the Htt protein as well its probable function. From this study, the CREBbinding protein & Multidrug transporter protein are found to be much similar in structural features & functional aspects of the Htt protein.

[more] The seed coat of the plant Borassus flabellifer L was extracted with cold water maceration. Well diffusion method was employed to determine the effect of antibacterial potential against Gram positive bacteria i.e., Staphylococcus aureus, Bacillus subtilis, Gram negative bacteria i.e., Escherichia coli, Klebsiella pneumonia and Proteus vulgaris, and Fungi Penicillium chrysogenum. Aqueous extract of the plant has showed inhibitory activity on different bacterial species and fungal agents employed against standard antibiotic drugs like Penicillin, Erythromycin and Griseofulvin. The phytochemical analysis revealed that the aqueous extract found to possess saponins, flavanoids and steroids, which may involve in showing antibacterial and antifungal activity. The UV-Visible spectrum of aqueous extract of B. flabellifer seed coat extract, indicate that four peaks with maximum absorbance at 240 nm, 280 nm, 540 and 620 nm. Further isolation and characterization of putative compound is under progress.
