  I 'd like to add that if these mutations seem not to change the amino acid sequence, u can allow it ... But if u are going to have any truncation in ur amino acid sequence , in case where ur gene sequence codes for a stop codon, the issue must be addressed...

  Hi Neeraja, I would like to know if u do an agarose gel purification after the RE digestion. With my experience what I noticed is that prolonged exposure to UV light during the time of excising the band results in so many mutations. I think u can have a check over it . And I wud like to know if u do a double digestion to ur plasmid and vector... Regards, Aishwarya Sriraman

Since there are many alkaloids, may i know from which plant species you are trying to extract the alkaloid... Hope I might help you ...
