Varital purity analysis through SSR marker

I am goining for genetic purity test of our hybrid chilli (capsicum annum) through SSR marker technique. please help me for the procedure for that. such as how sceern the primer and primer desiging? how select the best primer pair for pcr? It is possible to find out the best primer online only or need to screen practicaly in lab.

    Primers must be available in any paper, usually do you select in base of their PIC (polymorphic information content), if it's close to 1, it's most polymorphic, so coincidences between samples must be more significative. There's another factor that will be a problem that is the quality of PCR product (it's very important if you're checking in Polyacrilamide Gels) because in some primers do you see spots, not a well defined band... too bad that almost never information about quality is attached... it's a problem to find little differences in Polyacrilamide gels, other way is using primers marked with fluorophores and Automatic Sequencer

    If SSR is too problematic, you can always try AFLP or RAPD!

  • @ Swapna.. I will go with Giovanni It's good suggestion go with it but I have some following doubts and suggestion...
    1.Are you going for Parental purity or Hybrid purity????
    2. SSR markers also good source but they are less in nor available publicly....
