It is isolated from marine sample and I got through gram staining.

    As your mention, you got sample trough gram staining. Its look like gram positive bacteria, rod shape/ bacilli group. Should be try to isolate in Marine medium then firstly characterize with biochemistry test, follow to Bergy's Manual of Determinative Bacteriology, second sequencing its. If its not any organisms, its may be some debris that contaminated in your sample.

    This marine organism may have a pleomorphic character under microscopic examination.So it's not a confirmatory test, use biochemical identification tests or API kits to standardize your results. For more clarification and confirmation using DNA sequence. However be sure that this microscopic sample related to your final results. Also use a freshly prepared sample to be examined.

    please mention the sampling location, surface waters/shore waters, nearer to rocks/sediments and tidal or intertidal, whether you have checked the fresh sample or not. it could be gram positive, but what is the magnification. if you can give this information, i can help you. Since i have gone through different bacterial, algal species for my doctoral work, i am keen to help you

    mam i preserved my culture in fridge . After few days again i did grams staining and found some cocci mixed wit rods . So now i am trying to make pure culture.
    By doing quadrant streaking will i get back that organism ? ....
    After this i will give for sequencing

    i got tat sample from thiruvanmur beach , Chennai . i dint go deep in to sea and collect , i just collected outwardly . its just water. then i did serial dilution and did spread plate on MRS agar medium. after 48 hrs of incubation i got this organism. it was round whitish and rough . it is gram positive rods .. rods were very long.

    oh yah surly i will do it . but recently i found tat in my culture there is a mixture of other organism .. it showed cocci wit rods . so by dong Quadrant streaking will i get back my culture ?

After serial dilution i did spread plate on MRS agar medium. after 48 hrs of incubation i got round , whitish , rough colony . by doing gram staining i found its gram positive rods .. those rods were very long. and its magnification was 100x .

    go back to the sea , collect a new sample and add to selective media ( add NaCl for selections for marine cultures) for: algae ( no sugar, only salts), streptomyces ( with antobiotics) bacteria, fungi (PDA), them take a new picture! :)

    if it's green in color, could be an algae...
    MRS AGAR is for LACTOBACILLI culture

    The Gram's reaction indicates it as a Bacillus sp.with spores.The cocci you are mentioning might be the spores of this Bacillus.In a slight older culture this things happens. The spore may be confused as Cocci.You can conduct a spore staining of the and I think your doubt will be cleared.

    Hi, analyse all the biochemical test and then confirm, what it is, simply who did gram staining and asked what it is? means, what i will tell, do full of tests and then confirm with bacteriology manuals

    i actually used MRS agar medium for bacteriocins .. but i guess tat org which i observed may be actenomycetes not sure ..
    and yah i vl do again wit fresh samples .

    ok you can try any streptomyces medium with antibiotics, try Novocin as a selective agent.

    there are also ITS 16s primers for bacteria, it would be faster, probabaly, to do a PCR and sequencing to identified you bacteria
