    My favorite tool is an old sofware that runs in mac OS named amplify, but is hard to get it, my second option is the program AnnHyb that is free, and there is other serial cloner.

    It depends on what you want to do. For in silico cloning, molecular applications I'd suggest Geneious, but you have to buy a license for the Pro version. pDRAW32 from Acaclone is my second option, it's free and very easy to use with useful functions. Again, it all depends what you want to do.
